Objective: To prepare a mouse cell model of HIV-1 cross-species rapid susceptibility and virus efficient replication.This mouse cell model could be used to prepare HIV-1 infected murine animal models for the purpose of achieving HIV-1 cross-species murine animal model infection studies and promoting these studies such as HIV-1 antiviral drug screening and vaccine preparation,etc.Methods: First,hCD4/hCCR5/hCyclin T1 lentiviral was used to infect murine leukemia cell line L1210 to achieve the receptor hCD4,co-receptor hCCR5 and tat protein driving factor Cyclin T1 required for HIV-1 infection in L1210 cells.Next,positive clone L1210 cells were infected with HIV-1,then HIV-1 RNA in cells and culture supernatants was tested to verify the susceptibility and virus replication of positive clone L1210 cells to HIV-1.Finally,CEMx174 cells were infected with cell culture supernatants to make it clear that the progeny virus is active virus with infection ability.Results: After L1210 cells infected with hCD4/hCCR5/hCyclinT1 Lentiviral,hCD4,hCCR5 and hCyclin T1 were highly expressed in L1210 cells.After L1210 cells infected with HIV-1,HIV-1 RNA was highly expressed in L1210 cells and cells culture supernatants.After CEMx174 cells infected with cells supernatant,HIV-1 RNA was highly expressed in CEMx174 cells.Conclusion: A cross-species murine cell model of HIV-1 susceptible was constructed successfully with receptor hCD4,co-receptor hCCR5 and the transcription driving factor hCyclin T1 highly expressed,which enabled HIV-1 rapid infection,virus efficient replication and secretion active virus.It made a good foundation for the following study in murine animal model of HIV-1 cross-species infection,and provided a new direction for these studies of the pathogenesis of HIV/AIDS,screening of antiviral drugs and preparation of vaccines.
