Experimental Study On The Implantation Of Beta Tricalcium Phosphate Combination With The Peptide That Specifically Bind To Osteoblasts In Rabbit Tooth Extraction

Tooth sites preservation technique(ARP)refers to the tooth during or after tooth extraction,take reduces alveolar bone absorption in order to save the hard and soft tissue dissection form and can promote the new bone formation of Alveolar bone.Bone tissue engineering(BTE)application in vitro culture of tissue engineering has biological activity organization,then transplanted tissues in the body,to restore,maintain,or improve the defect in the organization and its reconstruction function.Osteoblast specific polypeptide(its amino acid sequence is VLAVPWSEPGYL)is a weakly hydrophilic polypeptide structure with good biocompatibility.This experiment will beta tricalcium phosphate and osteoblast specificity polypeptide composite implanted rabbit tooth socket,to evaluate the osteoblast specificity polypeptide in tooth nest sites preservation of osteogenesis function,so as to provide new methods for site preservation techniques and the area planted to the problem of shortage of bone mass to provide new ideas and methods.The details are as follows:Part ? The preparation of beta tricalcium phosphate(?-TCP)/osteoblast specific peptideThe polypeptide powder was dissolved by dimethyl sulfoxide(1ul/mg)as solvent,and the mass concentration of polypeptide with deionized water was 160mg/ml,which was sterilized by 0.22 um filter membrane on the ultra-clean working stage.Will beta TCP scaffolds into osteoblast specific peptide peptides,mild concussion to ensure osteoblast specificity polypeptide peptide solution fully adsorption to the beta TCP, room temperature for 24 h,embedded tooth socket defect area.In the scanning electron microscopy,tricalcium phosphate the pores were interpenetrating with each other.Part ? Establishment of animal model of preservation defect at the loci of extraction pit of rabbit tooth.27 New Zealand rabbits were established to preserve the defect model of tooth extraction nests in the right lower jaw.In the randomized group,the experimental group was implanted with the specific polypeptide complex of tricalcium phosphate/osteoclast,and the material group was implanted with tricalcium phosphate(bone powder),and the blank group was not implanted with any materials or drugs.4,8 and 12 weeks after surgery,CBCT scan,X-ray scan and HE staining were used to detect the osteogenic effect.The results showed that: 1 :4,8,12 weeks after X-ray measurement of residual alveolar ridge relative length(residual alveolar ridge relative length = tooth extraction side length of residual alveolar/non tooth extraction side length of residual alveolar): the experimental group residual alveolar ridge relative length is greater than the materials and the blank group,materials is greater than the blank group,and the comparison between groups with statistical significance(P < 0.05).2 : Reconstruction of the cross-section of living CBCT in 4,8 and 12 weeks after surgery can be seen: as time passes,the materials of the experimental group and the control group gradually degrade,the new bone mass increases,and the bone density increases.At 12 weeks,the materials of the experimental group were completely degraded,and tooth extraction was basically repaired,and the newborn bone was basically fused with the surrounding bone.In the control group,there was still a small amount of undegraded scaffolds,and the newborn bone density was lower than that of normal bone,but it was better than the blank group.3.The morphological observation was observed in 4,8 and 12 weeks after surgery: the osteoblasts in the experimental group BBB 0 were more active,and the new bone mass was significantly stronger than that of the materials group and the blank group.Conclusion Osteoblast specific peptides can promote bone regeneration and repair of tooth extraction nests in rabbits,and achieve the aim of preservation of tooth extraction.
