Objective:Through the detection of expression level of T lymphocyte and related cytokines in peripheral blood of patients with myelodysplastic syndrome(MDS)and acute myeloid leukemia(AML),to study the what's the difference between the two diseases of the immune status and influence on its development.Methods:Cytometric bead array(CBA)and flow cytometry were used to measure the expression levels of Treg,IL-2,IFN-?,TNF,IL-4,IL-6,IL-10,IL-17 in peripheral blood from 20 patients with MDS [MDS group: 10 with RA and 10 with RAEB],15 patients with AML(AML group)and 10 healthy individuals(normal control group),and indirect evaluation each T lymphocyte immune status.Results: Compared with the normal control group,the levels of TNF,IL – 17,IL –2 and IFN-? in the RA group were significantly up-regulated(all P <0.05),the levels of IL-10 were significantly down-regulated and the levels of Treg?IL-6?IL-4 had no difference between normal control group and RA group.Treg?IL-10?IL-6 and IL-4 levels in patients with RAEB and AML were considerably up-regulated(both P <0.05)with TNF were down-regulated,and IL-17?IL-2?IFN-? levels had no difference compared with normal control group.The expression levels of TNF,IL-17,IL-2 and IFN-? in the MDS-RAEB and AML groups was significantly lower whereas the level of Treg?IL-10?IL-6?IL-4 was significantly higher than those in the MDS-RA group(all P <0.05).Conclusion:The immune status of T lymphocytes were differed among various stages of MDS and AML.Increased negative regulation factors in the low risk MDS lead to apoptosis with bone marrow hematopoietic function failure,while increased immunosuppression ? immune escape and tumor cells in the high risk MDS and AML lead to normal hematopoiesis dysfunction.Therefore,the immune system participates in the development of MDS,and we need to pay attention to the value of immunoregulation therapy.
