Objective:To investigate the effect of ?-lapachone on the proliferation and apoptosis of hpMan gastric cancer SGC-7901 cells by regulating the expression of nicotinamide phosphoribosyltransferase(NAMPT)gene.Methods:1.Use of NAMPT to specifically inhibit FK866 inhibits the expression of NAMPT in gastric cancer SGC-7901 cells.The viability and apoptotic capacity of SGC-7901 cell lines before and after inhibition of NAMPT were observedWestern blot was used to detect the expressioin of NAMPT protein.According to Western blot results,the appropriate inhibitor concentration was screened by analysis of variance.3.The MIT assay detects the IC30 of ?-lapachone on gastric cancer SGC-7901 cells.4.The gastric cancer SGC-7901 cells were divided into 4 groups:FK866 group,(3-lapachone group,FK866+?-lapachone group and blank control group.Western blot was used to detect four groups of gastric cancer SGC-7901.Related cyclin changes.5.SGC-7901 cells were divided into two groups:FK866+ gastric cancer cells and gastric cancer cells(blank control group).MTT assay was used to detect the proliferation of gastric cancer SGC-7901 cells in FK866+ gastric cancer cells and gastric cancer cells.Results:1.MTT results showed that when the concentration of ?-lapachone was 4?mol/L,the survival rate of gastric cancer SGC-7901 cell decreased to 30%.The proliferation of gastric cancer SGC-7901 cells in the FK866+ gastric cancer cell group was significantly lower than that in the gastric cancer cell group SGC-7901 cells,with statistical significance(p<0.05).2.Western blot results showed that FK866 can significantly reduce the expression of NAMPT and was concentration-dependent.The expression of AKT/PI3K protein in FK866+?-lapachone group and FK866 group was significantly lower than that in other groups(p<0.05).Conclusions:1.FK866 can reduce the expression of NAMPT gene in gastric cancer SGC-7901 cell line.2.The decrease of the exprssion of NAMPT gene can enhance the inhibitory effect of?-lapachone on gastric cancer cells.3.NAMPT induces proliferation of gastric cancer SGC-7901 cells through AKT/PI3K signaling pathway and inhibits apoptosis of gastric cancer SGC-7901 cells.
