Objective:To explore the role of HMGB1 in the allograft rejection of mouse ovarian transplantation,and hope to find an effective target to provide ideas for future allograft rejection.Method:(1)The first part: Established the mouse ovary transplantation rejection model in the renal capsule,and the model divided into allograft group(Allo group),isograft group(Syc group)and control group(C57BL/6): Allogroup: Barb/c mice as donors and C57BL/6 mice as recipients;Sycgroup: The donors as same as the receptors were C57BL/6 mice,and blank control was untransplanted C57BL/6 mice.1 weeks after transplantation,the mice were killed and the serum,ovary,kidney and spleen tissues were taken.Histomorphology,immunohistochemistry and Western blotting were used to detect the changes of histomorphology and HMGB1 content in the grafts.(2)The second part: Made a model to block the effect of HMGB1 on the rejection of ovarian transplantation in mice.The glycyrrhizin treatment group: intraperitoneal injection of GL 200ul/;Normal saline treatment group: intraperitoneal injection of NS 200ul/.Two groups of mice were injected intraperitoneally with glycyrrhizin and saline,respectively,one day before transplantation,the first day,third days,fifth days,seventh days after transplantation.A total of 5 times,200 ul/ each time.1 weeks after transplantation,the mice were killed and the serum,ovary,kidney and spleen tissues were taken.Histomorphology and immunohistochemistry were used to detect histomorphology and HMGB1 content in blocking HMGB1 after transplantation.Result:(1)We made a successful establishment of mouse ovary transplantation under the renal capsule rejection model.The results showed that the morphology of ovary transplantation in follicular structure was destroyed,interstitial infiltration were filled with lymphocytes and neutrophils,and the fibrous tissue formed.Allo group was more obvious than Syc group.(2)Immunohistochemical results showed that HMGB1 was mainly expressed in the nucleus of the Syc group.While in the Allo group,HMGB1 could be expressed in nucleus,cytoplasm and extracellular matrix,indicating that HMGB1 in the graft moved from nucleus to cytoplasm and even to extracellular.Western Blot analysis showed that the expression level of HMGB1 protein in grafts increased after transplantation,while the expression of HMGB1 protein in allograft group was higher than that in homologous transplantation group.(3)The reduction of local HMGB1 expression in the grafts after blocking HMGB1 treatment,which could improve the degree of rejection in ovarian transplantation.Conclusion:(1)HMGB1 participates in the development of mouse ovarian transplantation rejection,and its increase can reflect the severity of rejection.(2)Blocking HMGB1 has a protective effect on ovarian transplantation rejection.(3)The level of HMGB1 may be a prognostic evaluation of allogenic ovarian transplantation and a new therapeutic target.
