Background and aim: Cadherin Related Family Member 5(CDHR5)positivity is significantly associated with a longer overall survival time in many tumors,but its role in HCC is unknown.we aimed to analyze the relationship between the expression of CDHR5 and Hepatocellular carcinoma(HCC).Method: The expression level of CDHR5 protein in HCC cell line and normal liver epithelial cell line was detected by Western Blot assay.Other 22 paired fresh frozen cancer tissues and corresponding adjacent normal tissues were examined by the same way.To assess the expression levels of CDHR5 protein in HCC tissues and corresponding adjacent normal tissues,22 paired fresh HCC samples and corresponding adjacent normal tissues(at least 2cm distant from the tumor edge)were divided into two parts.One was immediately frozen in liquid nitrogen after surgical removal and then were stored at-80°C,which was used to extract protein for Western Blot(WB)assay.The other was stored in formalin fixed solution(40% formaldehyde solution)and then buried by paraffin in reserve which was used for Immunohistochemistry assay.After collecting specimens,the postoperative pathological information of all patients was paid attention to.The specimens were used to study which confirmed of HCC by pathology.Otherwise,it would not be used as the experimental material in this study.Immunohistochemistry was performed to detect the expression of CDHR5 in 50 HCC clinical specimens with its adjacent normal tissues and Kaplan-Meier survival curve was used to explore the relationship between inequable expressions of CDHR5 in different HCC patients.Result: In this study,Hep 3B2.1-7 and HL-7002 cell lines and totally 22 paired fresh HCC tissues and corresponding adjacent normal tissues were detected by Western Blot assay.The expression levels of CDHR5 in Hep 3B2.1-7 cell line and HCC tissues were significantly lower than those in HL-7002 cell line and adjacent normal tissues.To evaluate the association of CDHR5 protein expression levels with clinicopathological parameters,IHC staining of 100 sections(50 HCC and 50 adjacent normal tissues)revealed that CDHR5 protein was mainly present on the membrane of HCC cells.The expression of CDHR5 in HCC tissues was significantly lower than that of the adjacent normal tissues.For survival analyses we have used Kaplan-Meier survival curve to explore the relationship between inequable expressions of CDHR5 in different HCC patients,The survival curve suggested that CDHR5 higher expression group showed a significantly lower risk of death than lower expression group Conclusion: 1,Compared with normal hepatic epithelial cells,CDHR5 was expressed low in hepatocellular carcinoma HEP 3B2.1-7 cells.2,the expression of CDHR5 protein was lower than corresponding adjacent tissues.3,CDHR5 expression,AFP and TNM staging were independent factors affecting the prognosis of HCC.And CDHR5 was protective prognostic factors for OS.AFP and TNM staging were negative.
