Improving the efficiency of pig's in vitro embryo production is very important to the field of science today,because pigs have high medical value for the model of xenotransplantation and embryonic stem cell research.Therefore,it is widely concerned to improve the production of in vitro embryos,however,because of a lot of external factors,the rate of in vitro embryo production in pigs lower than in body,a major reason is the in vitro maturation system on oocyte not well enough,so the matured oocytes quality is not good enough.The purpose of this study is to determine whether crocin as an antioxidant enhances the antioxidant capacity and the development rate of porcine embryos during the in vitro maturation of porcine oocytes.We try to add separately 0,300ug/ml,400ug/ml and 500ug/ml with 44h maturation.when supplement 400ug/ml that the maturation rate would highest that achieve 86.80%(P<0.01).Moreover,When supplement 500ug/ml crocin that the ROS concentration get the lowest(P<0.01).When supplement 400ug/ml crocin get the highest GSH concentration(P<0.01).On the antioxidant genes,when supplement 300ug/ml that SOD's mRNA expression get the highest(P<0.01)5 and supplement 400ug/ml that CAT's mRNA expression get the highest(P<0.05),and supplement 500ug/ml that GPx's mRNA expression get the highest(P<0.01).On the apoptosis-related genes,when supplement 500ug/ml crocin that the ratio of Bcl-2/BAX get the highest(P<0.05),when supplement 500ug/ml crocin that the mRNA expression of Caspase3 get the lowest(P<0.01).The cleavage rate reached the highest level of 62.50%when 400ug/ml was added(P<0.01).Our test results showed that the addition of 400ug/ml crocin on oocyte in vitro maturation rate and cleavage rate increased most effectively,and addition of crocin on the oocyte in vitro maturation that intracellular ROS concentration decrease and GSH concentration increase effectively,in addition to change of antioxidant and apoptosis-related genes in cell mRNA expression.
