Objective:Toxoplasma gondii is an intracellular parasite that parasitizes a variety of nucleated cell organisms including Ihumans.The main method of infection is through the oral cavity.After infection,T.gondii can flow with the blood and destroy the organs and tissues of the organism,causing zoonoses.Especially when the host's immune capacity declines can be fatal,T.gondii is opportunistic pathogens.At present,little drugs have been found to specifically prevent and treat the disease.Although traditional therapeutic drugs such as sulfadiazine and spiramycin can treat the disease,the effect is not very good and large doses of drugs may cause serious side ef:fects to the body.As a traditional Chinese medicine,Areca nut has the effects of insecticide,hydration,and stagnation,which can be used to treat infectious diseases caused by parasites such as hookworms,aphids,and worms.In this experiment,whether there are anti-T.gondii active ingredients in Areca nut.Hoping to find effective anti-T.gondii active ingredients in Areca nut.Methods:In this study,cell proliferation experiments in vitro and experiments with T.gondii infection in vivo were used to determine whether the effective components of Areca nut have anti-T.gondii effect,whether it can treat toxoplasmosis while protecting the host's liver and spleen,and whether it can increase the survival rate.In vitro experiments,MTT assay was used to determine the cytotoxicity of the active ingredients in Areca nut and the selectivity of its action against T.gondii;in vivo experiments,an acute mouse model of T.gondii infection was constructed to detect the number of tachyzoites in abdominal cavity.The number of mice was tested for liver and spleen index and their visual morphology.The levels of ALT and AST in the blood of the mice were detected.The levels of GSH and MDA in the liver were measured.The same model was used for 7 days treatment to observe the morbidity and mortality of the mice within 30 days.The differences in mouse indices between the experimental groups were compared.Results:The effective inhibitory effect of Areca nut on the growth of HeLa cells was dose-dependent,and the selectivity index of arecoline and arecoline hydrobromide in the effective components of Areca nut was significantly better than that of trditional medicine spiramycin in vitro;In vivo,the more polar active ingredients,the Areca total alkaloids and the arecoline hydrobromide all can effectively inhibit the proliferation of T.gondii in mice,and the treatment with Areca total alkaloids is superior to high-dose spiramycin,Therapeutic effects of arecoline hydrobromide are equivalent to high doses of spiramycin,and the hepatosplenic index,visual morphology,and survival rate of the mice after treatment with Areca total alkaloids are all superior to those of high doses of spiramycin;The indicators of after treatment witn arecoline hydrobromide were comparable to those of high-dose spiramycin.Conclusion:This experiment demonstrated for the first time that Areca total alkaloids and arecoline hydrobromide in the active ingredients of Areca nut were better low toxicity and high anti-T.gondii activity in vitro and in vivo compared with the positive control drug spiramycin.Protect mice liver and spleen well and extend the life span of mice.By studying the effective activity of Areca nut against T.gondii,new effective components for treatment of T.gondii were discovered,which provided a new theory for the treatment of T.gondii.
