Objective:The role of traditional Chinese medicine in animal liver injury model has been widely recognized by scholars,but a single drug has a slow onset and it is rarely used clinically.At present,the study on the anti-hepatic injury of traditional Chinese medicine at home and abroad,especially the study of the interaction and mechanism of action after mixing of various drugs is still lacking,which directly limits the application of traditional Chinese medicine in the treatment of clinical diseases.The purpose of this study was to investigate the effects of Curcumin and Ecological power element on acute and chronic hepatic injury,and whether the combination of the two drugs have synergistic effect on liver injury,and further explore the mechanism of its anti-hepatic fibrosis.Method:Part ?:To study the anti-acute hepatic injury effects of Curcumin and Ecological power elementOne week earlier,BALB/c mice Curcumin and 3 different concentrations of Ecological power element suspension were intragastrically administered.After the last intragastric administration,an acute liver injury model was established by intraperitoneal injection of 0.2%CC14 oil.Serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)levels were measured in each group of mice;hepatic HE staining of mice was performed to evaluate the anti-acute liver injury effects of Curcumin and Ecological power element.Part ?:To investigate the anti-hepatic fibrosis effect of Curcumin combined with Ecological power element by regulating Nrf2 oxidative stress pathway and its mechanismBALB/c mice were injected intraperitoneally with 20%CC14 oil twice a week for 6 weeks.The hepatic fibrosis model was established by intragastric administration of suspensions of Curcumin,Ecological power element,and co-administration groups.Serum levels of ALT,AST,superoxide dismutase(SOD),glutathione peroxidase(GSH-PX),and malondialdehyde(MDA)levels in mice were measured.Wolf scarlet dyeing evaluated the effects of Curcumin,Ecological power element,and co-administration groups on hepatic fibrosis.Real Time PCR Detects mRNA Expression of Nrf2 and GSH-PX in Liver Tissues of Mice and Explores Curcumin,to investigate the mechanism of anti-liver fibrosis by regulating the Nrf2 oxidative stress pathway by Curcumin in combination with Ecological power element.Results:Part ?:To study the anti-acute hepatic injury effects of Curcumin and Ecological power elementA single intraperitoneal injection of 0.2%CC14 oil can cause an acute liver injury model in mice.The expression of ALT and AST in serum of mice after CC14 injection for 24 hours is significantly higher than that in the normal group(P<0.01);liver tissue is stained by HE.The observation showed that the cells in the liver tissue of the mice showed a large amount of necrosis.Curcumin and Ecological power element significantly reduced the levels of ALT and AST in mice compared with the model group,and histologically reduced the degree of acute hepatocyte necrosis in mice.Part ?:To investigate the anti-hepatic fibrosis effect of Curcumin combined with Ecological power element by regulating Nrf2 oxidative stress pathway and its mechanismThe continuous injection of 20%CC14 oil can effectively induce the formation of hepatic fibrosis model in mice.The expression of serum ALT and AST levels in the model group was significantly higher than that in the normal group.HE staining in liver tissue showed a large number of hepatocytes.Flake necrosis,cell fat-like lesions,lobular structure disorders,and accompanied by infiltration of inflammatory cells,proliferation and spread of fibrous tissue in the vascular zone,fibrous tissue hyperplasia in the liver parenchyma;Curcumin,Ecological power element and co-administration groups can be significantly reduced(P<0.01)The serum levels of ALT and AST in mice significantly reduced the hepatocyte damage and inflammatory cell infiltration in mice.Sirnia scarlet staining showed that liver fibrosis in mice was significantly less than that in the model group.The levels of SOD and GSH-PX in serum of mice with Curcumin,Ecological power element and co-administration groups were significantly higher than those in the model group(P<0.01),and the levels of MDA were lower than those in the model group.Real Time PCR showed that the expression levels of Nrf2 and GSH-PX mRNA in the liver tissue of Curcumin,Ecological power element and co-administration groups were significantly higher than those in the model group(P<0.05).However,there was no significant difference in the serological test results between the combination group and the Curcumin group,and the difference in liver fibrosis was not significant.Conclusions:1.Both Curcumin and Ecological power element have anti-acute liver damage.2.Both Curcumin and Ecological power element have anti-fibrotic effects.3.The mechanism of anti-hepatic fibrosis induced by Curcumin combined with Ecological power element may be related to Nrf2-mediated oxidative stress pathway.4.The combined use of drugs failed to significantly enhance the anti-fibrotic effects of Curcumin.
