The Platycladus orientalis,a treasure,not only can be used as a desert greening plant but also its volatile oil can promote hair growth.As our society develops,more and more attention has been paid to the beauty of hair.Hair loss has seriously affected the appearance of people,so it is more important to prevent hair loss and make hair growth more safely and effectively.According to the reported literatures,the volatile oil of Platycladus orientalis leaves was taken as the research goal,and the basic mechanism of its pharmacological action was studied to explore its mechanism of action on hair regeneration.At present,the study on the hair growth promotion of Platycladus orientalis is only the description of apparent morphology,and the mechanism of its effect has not been further studied.This paper researched the effects of volatile oil from P.orientalis leaves on the expression profile of ?-catenin protein in hair follicle cells of mice,and determined the mechanism,by which volatile oil from P.orientalis leaves promotes hair regeneration.The study is listed as follows:(1)The subject taked Platycladus orientalis leaves as the research material.In this study,according to the Pharmacopoeia,steam distillation was used to extract about 200 ml of volatile oil from P.orientalis leaves.GC-MS analysis of volatile oil identified that the main constituents contain terpineol acetate,?-elemene,cedrol,?-caryophyllene,?-caryophyllene,and(-)-Germacrene D and so on.(2)Using C57BL/6 mice as the research target,the volatile oil from P.orientalis leaves was diluted to different concentrations for mice,and 5% minoxidil was used as a positive control.HE staining technique,NIS-Elements D software and SPSS 17.0.1 software were used to statistically compare the hair growth of mice at different treatment time(7 days,14 days,21 days,and 29 days),including the time of skin color changing in the treatment area,the time of new hair growth,the length of new hair,and the length of hair follicles and so on,to analyze the effects of volatile oil from P.orientalis leaves on hair regrowth in mice.At the same time,the optimal concentration of volatile oil was preliminarily selected.The results show that 10%and 20% volatile oil had good effects on hair regeneration in mice during the four treatment periods of 7 days,14 days,21 days,and 29 days.(3)Taking TGF-?1 protein and ?-catenin protein as the research objects,hair follicle cells that were applied for 0 days and hair follicle cells that were applied 7 days in blank group and20% volatile oil group of mice were subjected to immunohistochemistry(IHC).The results indicate that ?-catenin protein is mostly expressed on the cell membrane and TGF-?1 protein is expressed in cells.(4)Using ?-catenin protein as the research object,the expression profile of the protein in hair follicles of mice was analyzed by Western blot,Image J software and SPSS software.The experimental datas display that the expression of ?-catenin protein is up-regulated in hair follicle cells of mice on the 0th,7th,and 14 th days after treatments,and the ?-catenin protein expression is highest in group of 20% volatile oil extracted from P.orientalis.After 21 days of treatments,the expression of ?-catenin decreased,and the expression of the protein in the hair cells of the volatile oil group decreased but tended to balance.Then,our experiments make clear that the essential oil from P.orientalis leaves inducts the expression of ?-catenin protein in hair follicles of mice,which promotes the regeneration of hair follicles.
