Part one: Rapid quantification of liver fat content in non-alcoholic fatty liver disease based on 2-point Dixon methodObjective To evaluate the feasibility and accuracy of the 2-point Dixon method for quantitaftying the liver fat content(LFC)of non-alcoholic fatty liver disease(NAFLD)patients, with hydrogen proton magnetic resonance spectroscopy(~1H-MRS)as the reference standard. Methods A total of 42 volunteers were underwent by 1.5-T MRI Dixon liver volume scan and ~1H-MRS scan. Then, the scan sequences were sent to the post-processing workstation of Siemens to measure the liver content. Results The liver fat content measured by Siemens syngo MMWP VE40 B software in the Dixon fat component image was recorded as LFCDixon, and the average was(15.7 ± 8.7)%(the range was 6.5% ~ 25.8%).The fat content of the liver with 1h-mrs was recorded as LFCMRS,and the average was(15.0 ± 9.9)%(range of 6% ~ 26%).Pearson correlation coefficient was 0.925(P< 0.0001).With the Bland-Altman method,95.2% data points were within the 95% consistency range.conclusion2-point Dixon method can quickly and accurately quantify the liver fat content of non-alcoholic fatty liver patients.Part two : Quantitatively Analyze Liver Fat Deposition in new-diagnosed T2 DM Patients on Dixon MethodObjective To explore the liver fat content(LFC)and related factors in patients with the new diagnosed type 2 diabetes mellitus(T2DM).Methods A total of 42 T2 DM patients(observation group)were collected.(30 males and 12 females, with an average age of(40.0 ± 11.1))and 28 cases of healthy control group(18 males and 10 females, with an average age of(37.2 ± 10.2)).The fat content of hepatic right lobe(VII)in the two groups was measured and compared by the original 2-point Dixon method. We analyze the correlation between the liver fat content of the new diagnosed T2 DM and body mass index(BMI)and total triglyceride(TG).Results The liver fat fraction in the T2 DM group and the healthy control group were 10.02 ± 3.41% and 8.70 ± 2.62%,respectively,The difference was statistically significant(t = 1.706,P < 0.05).Compared with the control group,BMI,waist-to-hip ratio(WHR),fasting blood-glucose,(FPG),fasting C peptide,TG,and LFC were significantly increased in the T2 DM group(P < 0.05),There was no statistically significant difference between total cholesterol(TC)and low density lipoprotein(LDL-C)(P > 0.05).Conclusion The liver fat content of T2 DM group was higher than that in the healthy control group, and it was correlated with body weight, body mass index, waist circumference,waist-to-hip ratio, triglycerideand fasting c-peptide.
