The mortality rate of cardiovascular disease(CVD)ranks first in China,and its occurrence is affected by various factors such as the genetic alterations,environment,and life styles.The pathogenesis of CVD is complex and the efficacy of current single-target drugs is not ideal.Yixinshu Tablet,a Chinese patent medicine,has a significant therapeutic effect on CVD.However,due to the lack of research methods,its molecular mechanisms are not yet clear.In this study,we used a systems pharmacology method to analyze the molecular mechanisms of Yixinshu Tablet for the treatment of CVD.First,a database of 792 compounds of Yixinshu Tablet was constructed.105 candidate active compounds were obtained based on three pharmacokinetic parameters: oral bioavailability,drug-likeness,and intestinal epithelial cell permeability.Systematic drug targeting and weighted ensemble similarity models were used to predict the indirect and direct targets of these compounds,and 119 targets related to CVD were screened out.Based on the above results,we constructed compound-target,target-disease,and target-pathway networks to analyze the multi-component,multi-target mechanisms of Yixinshu Tablet for treating CVD.Finally,the “CVD Pathway” was constructed to reveal the effect of the active compounds on multiple pathways through the synergy of inflammation module,angiogenesis module and blood pressure regulation module.This study utilized the systems pharmacology analysis approach for elucidating the molecular mechanisms of Yixinshu Tablet in the treatment of CVD,which may provide a new idea for the research of traditional Chinese medicine and a new resource for the development of new drugs for CVD.
