| Background Primary liver cancer is one of the most common malignancies in the world.It ranks sixth of new Occurrence of cancers in the world every year.It ranks second in all causes of cancer deaths and it threatens human health.Cancer stem cells(CSCs)belong to a special cell subtype in malignant tumor cell population.They not only have the malignant characteristics of unrestricted proliferation of cancer cells,but also have the characteristics of rapid self-renewal and multi-directional differentiation.Hepatocellular carcinoma stem cells can not only induce the occurrence of liver cancer,but also can play a role in promoting tumor growth and metastasis,and it is also an important cause of liver cancer resistance to radiotherapy and chemotherapy and recurrence after treatment.X-linked inhibitors of apoptosis proteins(XIAP)are the most important tumor-associated proteins in recent years and they are important members of the Inhibitors of Apoptosis Proteins(IAPs)family.XIAP has low or no expression in normal tissues,and high expression in hepatocellular carcinoma.Studies have shown that high expression of XIAP leads to a variety of tumor cells including liver cancer and other conventionalchemotherapy drugs such as VP16 and doxorubicin and other novel types.Anti-cancer drugs such as TRAIL(TNF-related apoptosis inducing ligand)and other important molecular mechanisms to resist the induction of apoptosis.Therefore,XIAP has become one of the key targets for research in tumor molecular therapy.This article intends to study whether XIAP can regulate liver cancer stem cells,promote their self-replication and induce tumors and its long-term survival effects on patients.Explore new ways to regulate liver cancer stem cells in the treatment of liver cancer,and provide new ideas for improving the therapeutic effect of liver cancer.Object 1.To study the ability of XIAP to regulate the colony formation ability of hepatoma stem cells and the ability to detach and purify hepatoma stem cells into spheres.2.The correlation between XIAP and cancer stem cell marker molecule CD133 was analyzed by clinical samples;based on the TCGA database,there was a specific correlation between the expression level of XIAP in patients with hepatocellular carcinoma and the prognosis of patients.Methods 1.The effect of XIAP on the colony forming ability and the ability to form a ball of liver cancer stem cells was investigated by using small RNA(si RNA)interference technology to specifically inhibit the expression of XIAP.2.Hepatoma surgical specimens were collected and Western blotting was used to detect the expression of XIAP and tumor stem cell marker molecule CD133 in hepatocellular carcinoma.The correlation between XIAP and hepatoma stem cell marker molecule CD133 was analyzed.Based on TCGA database,hepatoma patients were studied.The relationship between the expression level of XIAP and the survival rate of patients with hepatocellular carcinoma.Results 1.After silencing XIAP gene with si RNA,the colony forming ability of hepatocarcinoma stem cells in vitro is weakened;and the ability of cancer stem cells to inhibit the ball formation is obviously inhibited.2.Western blotting analysis showed that the expression of XIAP was higher in HCC tissue,and the expression of tumor stem cell marker CD133 was also significantly increased,suggesting a correlation between XIAP and CD133 in liver cancer tissue;the comparison of XIAP gene expression with the survival rate of liver cancer patients.The overall survival time was significantly higher in the XIAP low expression group than in the high expression group.Conclusions 1.Silencing of XIAP gene by si RNA can inhibit the colony forming ability and the ability of forming the hepatoma stem cells.It is suggested that XIAP has a significant protective effect on the survival of liver cancer stem cells,and may be an important regulatory molecule that affects the survival and replication of liver cancer stem cells.2.XIAP is associated with cancer stem cell marker CD133 in hepatocellular carcinoma.XIAP may be an important regulator of liver cancer stem cells;detection of XIAP expression may have predictive value for survival of patients with hepatocellular carcinoma. |
