BackgroundBerberine is an isoquinoline quaternary alkaloid,one of plant-derived compounds,which can be abstracted from various kinds of herbal medical plants such as Berberis aristata,and Coptis chinensis.Nowadays,it has been utilised as traditional herbal medicine.Many researches demonstrate that berberine has plenty of pharmacological effects,which compose antioxudant and antitumor function.Apart from these,it can play an important role in treating against obesity and Alzheimer 's disease.In recent years,as scientific experiments are going on,berberine's anticancer effect is increasingly paid more attention by researchers.Studies demonstrate that berberine is capable of effectively inhibiting hepatic cancer,bone tumor and esophageal cancer.etc.However,there is less study concerning berberine's effects on renal cell cancer.Thus,it has certain value to be involved in this study.As is well known,DNA is a kind of long-chain polymer,carrying biological genetic information.Hence,DNA plays significant roles in all varieties of species.Many anti-tumor drugs can induce cancer cell DNA fraction through different kinds of mechanisms to restrain tumor.Nonetheless,cancer cells would immediately trigger DNA damage repair mechanisms,as DNA is broken down,which encompass homologous recombination repair(HRR)and non-homologous end joining(NHEJ).Consequently,halting cancer cells' DNA damage repaire is a prominent approach to keep tumor from progressing.Objective To determine the effects of berberine(BBR)on the growth,cell apoptosis and DNA damage and repair of human renal cell carcinoma(RCC)cells.MethodsRead articles relating to berberine's anticancer effects in various kinds of tumors in order to determine study of berberine's effects on renal cancer cell.The effects of berberine on the proliferation of A498 and 786-0 RCC cells at concentrations of 0-240 ?mol/L were tested with CCK-8 testing kit.Based on the results of CCK-8 test,RCC cells were divided into 3 groups,control group(0 ?mol/L),30 ?mol/L group and 60 ?mol/L group.The cell apoptosis after BBR treatment was evaluated with Annexin V-FITC/PI Apoptosis Detection Kit.The expressions of proteins involved in the apoptotic pathway and DNA damage repair pathways,including pro-caspased3,cleaved-caspased3,yH2A.X and Ku70,were assessed with Western blotting.Results1.The proliferation of RCC cells was significantly suppressed by BBR in a time and dose dependent manner.2.Cell apoptosis was obviously increased after berberine treatment at an optimal concentration and time point.3.Western blotting underpined the previous finding that berberine co?ld increase renaLcancer celLapoptosis.Besides,berberine could induce renaLcancer cell DNA damage and suppress cell DNA damage repair system,then cell growth could be successfully inhibited.Study showed that,in comparison with the control group,the 30 and 60 ?mol/L groups showed increased expressions of cleaved-caspase3 and yH2A.X,but decreased expression of Ku70.Compared with the 30 p.mol/L group,the 60 ?mol/L group showed increased expressions of cleaved-caspase3 and ?H2A.X,but decreased expression of Ku70.ConclusionOur study demonstrates that berberine can effectively suppress renal cancer cell proliferation,induce renal cancer cell apoptosis and DNA damage,and inhibit DNA damage repair.
