OBJECTIVE: To explore the effect of ginsenoside Rg1 on the transdifferentiation of astrocytes into neurons..METHODS: First,preparefor Sprague-Dawley adult rats model of spinal cord injury,andthen cut the rat's spinal cord after a week,At last,culture the cells in ACM(astrocyte conditioned medium)after digestion and centrifugal.Here the experiment is divided into two groups,one group putting ginsenoside Rg1 into the ACM for culturing together,another group is set to blank contrast,after a week cells being formed into the ball,then join the differentiation of neural stem cells in the medium by nature,inverted microscope to observe the cell growth in the two groups,The expression of nestin and GFAP in the two groups and the MAP-2 positive expression of the cells were identified by immunofluorescence assay.The differences in expression of nestin and MAP-2 were detected by Western Blot.Results: After a week there are many astrocytes spheres in two groups,and astrocytes spheres were coexpress GFAP and nestin by immunocytochemistry,and after induction of astrocytes spheres to differentiate in two groups,some cells can show MAP-2 positive expression,but the two groups in the MAP-2 positive expression rate is different,Rg1 groups is(4.39 ±1.00)%,while the control group(2.63 ±0.76)%,statistical test P < 0.05.Western blot analyze the expressing of nestin and MAP-2 in different time,Rg1 can promote the expression of nestin after the injury of astrocytes,and promote the expression of MAP-2 after differentiation.CONCLUSION:Ginsenoside Rg1 can promote the transdifferentiation of astrocytes into neurons and provide a new way for clinical treatment of spinal cord injury.
