On the basis of previous studies on chemistry,pharmacodynamics and pharmacokinetics,we used LC-MS technology as the focus of secondary metabolites in vivo.The metabolic behavior of each component was explored,and the in vivo metabolism of the chemical constituents of the effective parts of Caulophyllum robustum Maxim was studied in depth.After taking the effective parts of peony,the blood,bile,urine and fecal samples were collected at different time points.In order to isolate and identify the metabolites of the effective parts of the tree peony in vivo,the possible biological transformation process in the body is speculated by using chemical,chromatography and mass spectrometry.At the same time,the metabolic transformation pathways of these metabolites are described.Finally,we hope to be able to explain the 'unique mechanism of the effective parts of Paeonia suffruticosa.In order to investigate the in vivo transformation of the effective parts of rheumatoid arthritis(RA)in Peony(RA),this paper studies the metabolic behavior of four saponins and two alkaloid components in the effective parts of the tree peony.Analysis of the metabolic transformation of components of similar substances and similar structures in vivo.Firstly,mass spectra of three terpenoid saponins and alkaloids were systematically analyzed by negative ion mode.Secondly,the metabolic mechanism and metabolic process of the drug effect active substance in the body are discussed,and the mass spectrometric cracking behavior of the compound is combined with the possible metabolic characteristics.Using the effective method of characteristic ion extraction,we can quickly identify and identify prototypes and metabolites from complex biological samples.Finally,the pharmacodynamic material basis of the effect of three kinds of terpenoid saponins and alkaloids on RA effective parts of peony tree peony was established.The main contents of this paper are as follows:(1)Mass spectrometric fragmentation of three terpenoid saponins and alkaloidsAccording to the structural characteristics of compounds in Paeonia suffruticosa,the cleavage behavior of various components was deduced.In the negative ion mode,Cauloside C break will produce m/z 765,603 and 471 characteristic ion fragments;Cauloside D break will produce m/z 1073,765,603,and 469 characteristic ion fragments;Leonticin D contains a side hydroxyl group,so it will produce m/z 1089,927,619 and 469 characteristic ion fragments;Cauloside H break will produce m/z 1251,781,619 and 469 characteristic ion fragments.Magnoflorine were found to produce m/z 325,310 demethylated ion fragments,m/z 322 dewatered ion fragments,and m/z 295 characteristic ions formed by loss of 45 Da amino groups through mass spectrometry data.Caulophylline B mass spectrometry data were found to produce m/z 327 and 312 demethylated ionic fragments,as well as m/z 297 characteristic ions formed by the loss of 45 Da amine groups.There are many similar components in peony leaves,and similar characteristics are found in mass spectrometric cleavage behavior.(2)In vivo metabolism of three terpenoid saponins from effective parts of Caulophyllum robustum MaximIn this chapter,liquid chromatography was used to analyze the biological samples of different sampling time points in rats,and to discover and deduce the possible structure of the prototypes and metabolites from the complex biological samples.Seven prototype components and eight metabolites were found in rat plasma,including desaccharides,saponins and glycine conjugates.No prototypical components were found in bile.six metabolites were found,including disglycosyl products,sulfides and glucuronide conjugates.Five prototypes and eight metabolites,including disglycosyl products,saponins,sulfides and glucuronic acid conjugates,were found in urine.Four prototypical components and nine metabolites were found in feces,including desaccharide base products,saponins,sulfide and glucuronic acid conjugates.To sum up,the same metabolites are found in these different biological samples,suggesting that glycidyl,hydroxylation,glucuronization,sulfation and glycine binding may be the important metabolic pathways of three terpenoid saponins in rats.(3)In vivo metabolism of alkaloids from the effective parts of Caulophyllum robustum MaximIn this chapter,liquid chromatography mass spectrometry(hlc-ms)was used to analyze the biological samples and blank biological samples.Three prototyping components and eight metabolites including demethylation and hydroxylation were found in rat plasma.Four prototypes and six metabolites,including demethylation,were found in bile.There prototype components and five metabolites,including demethylation,hydroxylation and acetylation products,were found in urine.There archetypal components and five metabolites including demethylation and demethylation products were found in feces.In conclusion,in these different have the same metabolites in biological samples,found that prompt,methyl,hydroxyl and sulfate,may be alkaloids component important metabolic pathway in rats.This study conjectured the metabolic characteristics of the components in the effective parts of the tree peony,and obtained the overall information on the biotransformation of the effective parts of RA,which provided an effective way to improve the efficiency and the discovery of active components in the basic research of Chinese medicine.This study clarified the metabolic pathways of the main active components in the effective parts of peony,and further elucidated the anti RA substance basis of peony,and provided scientific information for the discovery of the new effective components.
