With the rapid development of society,the pace of life continues to accelerate,the pressure people are facing from all aspects also increased.Men,not only to face the pressure from the community,but also bear the responsibility of the family,so they tend to bear greater psychological stress than other groups.Psychological stress has become one of the main factors affecting male fertility,psychological stress can damage the spermatogenic epithelium,causing germ cells and sertoli cell gap expansion,damaging the cell structure,causing nutritional deficiencies,resulting in spermatogenesis obstacle and reduced androgen synthesis.The study found that psychological stress will activate the Fas signaling pathway in the ovary and induce apoptosis to damage the development of oocyte.However,it is unclear whether psychological stress will directly affect germ cells by activating Fas signaling pathways.To this end,we used the restraint stress model to simulate the psychological stress,the male mice for 48 hours in a row,by detecting the fertilization rate,the number of births,sperm motility and apoptosis index of spermatogenic tubules,to study the role of Fas signaling pathway in the process of restrain stress damaging germ cells.And further verify the role of Fas signaling pathways through gld mice to explore the specific mechanisms of injury to germ cells by restraint stress,to help us better understand the mechanism of psychological stress on male fertility.Our results show that:1.Male mouse suffered continuous restraint stress for 48 hours caused the in vitro fertilization rate lower than normal mouse.2.Male mouse suffered continuous restraint stress for 48 hours,led to decreasing in mitochondria membrane potential of spermatozoa.the ratio of lower mitochondria membrane potential of spermatozoa is higher than normal spermatozoa.3.Male mouse suffered continuous restraint stress for 48 hours,the vitality of sperm decreased faster than sperms of mouse untreated in the in vitro preservation process.4.The expression of Caspase-3,Fas and FasL was up-regulated in Kunming mouse and C57BL/6J mouse seminiferous tubles.Restraint stress can cause apoptosis of seminiferous tubules through Fas / FasL signaling pathway.5.Male mouse suffered continuous restraint stress for 48 hours,the expression of Caspase-3 was up-regulated in gld mouse,Fas/FasL signal pathway is useless in gld mouse,some other pathway caused apoptosis in gld mouse.
