Objective:Chinese medicine theory of"liver catharsis,spleen operation"can be used to explain the higher incidence of Hyperlipidemia,Sini powder can therefore be chosen as a drug to treat high cholesterol.Study of Sini powder on hyperlipemia C57BL/6J and lipid metabolism in mice and its mechanism,and provide evidence for clinical treatment of hyperlipidemia.Methods:Select 40 SPF C57BL/6J mice as a research object,using random number tables divided into normal control group,the Group of high-fat,high-fat plus Sini powder(1.5mg·g-1·d-1)intervention group,high-fat plus aurantii(1.5 mg·g-1·d-1).The normal group was fed normal diet and the others were fed with high fat diet.The intervention group,respectively,saline,Sini powder decoction,aurantii decoction administration by gavage.Experimental intervention after 90 days weighing pick eye blood,liver and small intestine,respectively.Biochemical parameters using automatic biochemical Analyzer in detection of serum triglyceride and high-density lipoprotein cholesterol,low-density lipoprotein cholesterol levels,A-I ELISA method for detection of serum apolipoprotein level and measured by real-time fluorescence quantitative PCR reaction in liver and APOA-I mRNA gene expression in intestinal tissue.Results:1 Weight:Sini mice,aurantii group body weights of mice compared with mice Hyperlipidemia model set weight decreased variance?P<0.05?,high fat mice body weight increased significantly relative to normal weight group,the difference was statistically significant?P<0.05?.Sini mice body weight more slowly.2 Biochemical parameters:High fat group compared with control group mice serum TG and LDL-C average increased significantly?P<0.05?.Compared with the high fat group,Sini mice and bitter orange mouse serum LDL-c and TG average was significantly reduced?P<0.05?.Compared with the high fat group Sini serum HDL-c levels were significantly higher in mice?P<0.05?,the elevation of serum HDL-C level in mice of aurantii group,the difference was statistically significant?P<0.05?.Sini group compared with mice with Fructus aurantii immaturus group TG,LDL-c water decreased and HDL-c levels rise,there was a statistically significant difference?P<0.05?.Sini APOA-I content in the serum of mice was significantly higher than the rest of the study group?P<0.05?,citrus aurantium APOA-I content in the serum of mice than in the high fat Group?P<0.05?.3 The relative expression of APOA-I mRNA in liver and small intestine:In the liver tissue,Sini group expression of APOA-I mRNA is about 5 times the high fat group was Fructus aurantii immaturus group APOA-I mRNA expression was 1.5 times significantly?P<0.05?.Sini groups in the intestinal tissue of expression of APOA-I mRNA expression was most high,about 2.4 times the high fat group,twice times was Fructus aurantii immaturus differ significantly?P<0.05?.Sini group,Orange group,normal liver APOA-I mRNA expression levels were higher than in the small intestine.Conclusion:1 Sini Powder can prevent hyperlipidemia by clearing Yang,resolving depression,soothing the liver and regulating spleen2 Sini experimental mice can significantly reduce weight,reduce hyperlipemia mice serum TG level and increased HDL-c,LDL-c levels.3 Sini hypolipidemic effect and the mechanism is effects the liver and small intestine increased secretion of APOA-I mRNA gene expression of APOA-I,thereby affecting HDL levels,enhance the reverse transport of cholesterol in serum and excluded.
