Objective To investigate the effect of hypertonic glucose joint injection on synovitis caused by sodium monoiodoacetate(MIA)in rats.Methods Thirty male rats weighing about 200 g were randomly divided into three groups:blank control group(group A),negative control group(group B)and hypertonic glucose group(group C).Rats in group B and C were treated with MIA(50ul/3ml)injection of knee osteoarthritis.On the 8th day after model establishment,group C rats were injected intra-articularly with 12.5% hypertonic glucose(50 ?l).Group B rats were injected intraperitoneally with normal saline(50 ?l)and injected once a week for four weeks.Group A was a group of healthy rats.Nothing is done.Before and 36 days after modeling,the general conditions(mental state,activity,body weight,appearance,etc.)of the three groups of rats were observed.Before and 14 d and 36 d after modeling,the rats in the three groups underwent Paw withdrawal threshold(PWT)test.At the36 th day after modeling,the levels of TNF-? and IL-1? in the synovium of the three groups of rats were measured by Western-blot,and the gross observation and pathological scores of the femoral condyle were performed.Results(1)On the 36 th day after modeling,the general observation of the three groups of rats was compared with that of group A.The fur of group B and group C were disorderly brown;The body weights of rats in group B and group C were significantly lower than those in group A(p<0.05),and those in group C were higher than those in group B(p<0.05);The knee joint mobility in group B and group C significantly decreased(p<0.05),and group C was higher than group B.(2)On the 14 th day after modeling,the 50% PWT value of group B and group C rats was significantly lower than that of group A(p<0.05).At the 36 th day after modeling,50% PWT in group C rats was lower than normal group,but higher than50% PWT in group B rats(p<0.05).(3)On the 36 th day after modeling,the levels of TNF-? and IL-1? in the synovium of group B and group C were significantly higher than those of group A(p<0.05),and group C was lower than group B.Group(p<0.05).(4)At the 36 th day after modeling,the gross scores of rat cartilage in group B and roup C were significantly higher than those in group A(p<0.05).There was no significant difference between the two groups(p>0.05).(5)At the 36 th day after modeling,the structure of articular cartilage in group A was clear.In group B,the structure of articular cartilage was disordered,only a few normal cartilage cells were seen,a large number of fibrocartilage was formed on the surface of cartilage,and the surrounding cells proliferated severely,and the cell matrix disappeared;In group C,the cartilage layer became thinner,cartilage cells were significantly reduced,and a small amount of fibrocartilage was formed.Mankin,s scores in group B and C were significantly higher than those in group A(p<0.05),and group B was higher than group C(p<0.05).Conclusions(1)Intra-articular injection of 12.5% hypertonic glucose can reduce the protein expression levels of TNF-? and IL-1? in synovium.(2)Intra-articular injection of 12.5% hypertonic glucose has a repairing effect on rat cartilage.
