Effects Of Panax Notoginseng Saponins In Combine With Cisplatin On Growth,Metastasis,and Apoptosis Of HepG2 And SMMC-7721 Cells

Objective:To investigate the effects and the mechanism of PNS in combine with DDP on the HepG2 and SMMC-7721 cells.Methods:The cellular proliferation of HepG2 and SMMC-7721 cells treated with PNS(200?mol/L)or/and DDP(5?mol/L)at the indicated concentrations was detected by MTT assay;The migration and invasion of HCC cells was detected by wound healing assay and matrigel invasion assay;Flow cytometry assay was used to analyze the changes of cell apoptosis;The protein expression was measured by western blot assay.Results:PNS and DDP would inhibit cell viability of HCC cells,in a dose-dependent manner.PNS in combine with DDP significantly inhibited the cell growth,migration and invasion of HCC cells.Besides,PNS in combine with DDP significantly inhibited the expression of Bcl-2,and promoted the expression of Bax to promote the apoptosis of HCC cells.Conclusion:In constract with the effect of inhibiting by PNS and cisplatin alone,the cell growth,migration and invasion of HCC cells can be observably inhibited by PNS in combine with DDP.Inhibiting the expression of Bcl-2,and promoting the expression of Bax,PNS in combine with DDP promotes the apoptosis of HCC cells.
