Objective:To study the effect of schisanhenol on the proliferartion of Cardiac Fibroblasts induced by Angiotensin ?.and to explore the mechanism of Schisanhenol in treating Myocardial Fibrosis and the effects of Connexin 43,TGF-?1.Methods:SD neonatal rat heart was used in this study and hearts were digested with 0.1%Type ? collagenase digestion,then purified Cardiac Fibroblasts were obtained by the methods of digestive enzyme and differential adhesion;Cardiac Fibroblasts was identified by immunofluorescence assay;The effect of Schisanhenol on proliferation of myocardial fibrosis was observed by Methyl Thiazolyl Tetrazolium(MTT)assay.By means of Western Blot metheds,the levels of TGF-? 1,Connexin 43,Collagen ? and Collagen ? proteins were examined by Angiotensin II and Schisanhenol drug intervention.Results:1.MTT experimental results showed that Ang ? 10-7mol · L-1 group has significant difference compared to control group and on the proliferation of Cardiac Fibroblasts obviously(P<0.01);Within a concentration coverage,Schisanhenol inhibited on the proliferation of Cardiac Fibroblasts induced by Angiotensin ? in a dose-dependent way(P<0.05,P<0.01,respectively).2.Western Blot experimental results showed that Ang ? 10-7 mol · L-1 group has significant difference compared to control group and enhances the protein expression of Connexin 43 obviously(P<0.01);Within a concentration coverage,Schisanhenol inhibited the protein expression of Connexin 43 in a dose-dependent way(P<0.05,P<0.01,respectively).3.Western Blot experimental results showed that Ang II 10-7mol · L-1 group has significant difference compared to control group and enhances the protein expression of collage ?,collagen ? and TGF-?1 obviously(P<0.01);Within a concentration coverage,Schisanhenol inhibited the protein expression of Collage ?,Collagen ? and TGF-?1 in a dose-dependent way(P<0.05,P<0.01,respectively)Conclusion:1.Ang ? induces proliferation of Cardiac Fibroblasts by enhancing the protein expression of TGF-?1,Collagen ?,Collagen ? and inhibiting the protein expression of Connexin 43.2.Schisanhenol can inhibit the proliferation of Cardiac Fibroblasts by enhancing the protein expression of Connexin 43 and inhibiting the protein expression of TGF-?1,Collagen ?,Collagen ?.
