Curcumin Exerts Anti-inflammatory Effect On LPS-induced Microglial Ocells Through PI3K/AKt Signaling

Abstract/Summary:
Aim:To study the protective effect of curcumin on LPS-induced BV-2 cell injury and its mechanism of action.Methods:The microglia BV-2 was divided into four groups:control group,LPS model group,10 μ mol/L curcumin group,20 μmol/L curcumin group,and 40umol/L curcumin group.The curcumin group was pretreated with 10 μ mol/L,20 μ mol/L,and 40 μ mol/L curcumin for 1 hour,and then treated with lg/mL LPS for 24 hours.The LPS group was treated with lg/mL LPS.Treatment for 24h;control group cells without any treatment,cells in 5%C02 37℃ incubator,logarithmic growth phase cells for experiments.Using MTT assay,cytotoxicity of curcumin on BV2 cells further showed cell viability among different groups;the production of NO in different cells was compared by Griess method.The expression of TNF-α,IL-1β and IL-6 cytokines was observed by ELISA.The expression of iNOS,NF-κB p65,p-AKT and other proteins were detected by Western blot.Results:1.MTT results showed that different concentrations of curcumin had no toxic effects on BV2 cells.2.Griess method results show that LPS can significantly increase NO production in BV2 cells,and curcumin can significantly reduce the production of NO after dose-dependent manner.3.ELISA results showed that LPS can significantly increase the expression of TNF-α,IL-1 β and IL-6 cytokines in BV2 cells,curcumin treatment can significantly reduce TNF-a,IL-1β and IL-6 in BV2 cells.Cytokine expression.4.Western blot results showed that LPS can significantly increase the expression of iNOS and NF-κB p65 and p-AKT protein in BV2 cells.Curcumin treatment can significantly reduce iNOS and NF-κB p65 and p-AKT in BV2 cells.Protein expression.Conclusion:1.Curcumin can significantly increase the survival rate of LPS-induced microglia and protect it.2.Curcumin decreased the apoptosis rate of LPS-induced microglial cells,and its mechanism was mainly through the activation of PI3K/AKT pathway to inhibit the activation of NF-kb,down-regulate iNOS,and reduce the production of NO.3.Curcumin can protect LPS-induced microglial cells by inhibiting the activation of NF-κB,reducing the expression of iNOS,NF-κB p65,p-AKT protein,and reducing the production of cytokines.
