Objective:Coronary heart disease is one of the most important cause of morbidity and mortality worldwide and atherosclerosis is a major factor in the pathogenesis of coronary heart disease.Its pathological characterized by chronic inflammation of blood vessels due to the accumulation of fatty plaques on the arterial wall,which eventually will lead to ischemia of heart and other organs.The main risk factors leading to coronary heart disease were hypertension,diabetes,hyperlipidemia,history of tobacco and alcohol,age,obesity and family history.Coronary heart disease is influenced by many factors,among which the most important one is the complex synergy between genetic background and environmental factors,therefore the study of gene level has become an indispensable part of the treatment of coronary heart disease.And the polymorphism of gene locus is more and more closely related to coronary heart disease.Based on the previous literature and experimental findings,it's found that the expression of cytokines associated with LTBP1 gene was higher in coronary heart disease patients.This study was conducted to study the difference of the distribution of LTBP1 gene locus rs2290449(C>T)'s genotypes between coronary heart disease group and healthy control group to study correlation of the polymorphism of LTBP1 gene locus rs2290449(C>T)and coronary heart disease.Methods:1)The experiment included 761 subjects,378 of whom were diagnosed with coronary heart disease,while 383 were healthy controls.2)Measures will be taken to ensure the exclusion of unrelated variables of the healthy control group on the results of this experiment.The blood samples of experimental subjects was used for the extraction of DNA.In order to measure the polymorphism of LTBP1 gene locus rs2290449,the DNA extract with concentration up to standard was sequenced by PCR-RFLP technique.The deteramined genotypes,the distribution of allele frequency and genotype in coronary heart disease group and healthy control group was analyzed by SPSS statistical software 22.0.The general data of coronary heart disease group and healthy control group were compared by X2 test and t-test.Whether P value is less than 0.05 is used to indicate whether it has statistical difference between the two groups.The data is tested by normal distribution and homogeneity test of variance.The normal distribution was expressed by mean ± standard deviation.The genotypic distribution was analyzed by Logistics regression model,and the relationship between genotype and risk of coronary heart disease is determined by estimating the odds ratio(OR)and the 95%confidence interval(Cl).Results:By comparing the general data between the coronary heart disease group and the healthy control group,it was concluded that hypertension history,diabetes history,drinking history,total cholesterol,triglyceride had statistically difference.The number of patients with hypertension,the number of patients with diabetes,alcoholics,the content of total cholesterol and triglyceride in coronary heart disease group are all higher than that in the healthy control group.Reference to TT genotype,CC genotype is a protective factor for coronary heart disease and the OR value is 0.521.Reference to CC genotype,the risk of coronary heart disease was 2.039 times and 1.918 times higher in CT genotype and TT genotype,respectively.C allele is a protective factor for coronary heart disease,which OR value is 0.691 times.Conversely,the risk of coronary heart disease in T allele was 1.447 times higher than that in C allele.Conclusion:The polymorphism of LTBP1 gene locus rs2290449(C>T)is associated with coronary heart disease.The risk of coronary heart disease is higher in people carrying T allele or TT genotype than in the general population.
