Background and purpose: Background:Preeclampsia,a pregnancy complication that is more common disease,one of maternal and perinatal mortality risk factors are also accepted by most scholars as.At present,the research on the causes of morbidity is more extensive,but the exact cause is different.Among them,as a kind of acceptance by the majority of scholars is the placenta injury,but also one of the main causes of preeclampsia.Among them,the cause of insufficient remodeling of placental spiral arteries during placental injury is one of the most widely recognized and important factors for induction.Vascular Endothelial Growth Factor Receptors-1(VEGFR-1),Soluble Vascular Endothelial Growth Factor Receptors-1(SVEGFR-1)and Vascular Endothelial Cell Growth Vascular Endothelial Growth Factor(VEGF)can be used as an important indicator to judge and measure the state of involvement of angiogenesis and endothelium damage.Long-chain non-coding RNA(1ncRNA)is known as an RNA transcript of non-coding proteins.The basic characteristics are summarized as follows: Found in eukaryotes,RNA that is formed by RNA polymerase II transcription,lacks a long reading frame,and is longer than 200 nucleotides,but often possesses the structural characteristics of mRNA.LncRNA-LOC391533 is 1434 bp in size and is a LOC391533 gene transcript located on chromosome 3p21.31.LOC391533 is the pseudogene 1(VEGF1P1)of fms-like tyrosine kinase 1 of the VEGF-1 gene.The VEGF-1 gene is located on chromosome 13q12.VEGF has four receptors: FLT-1,KDR,FLT-4,NRP-1,while sVEGFR-1 is a soluble form of VEGFR-1,and VEGR-1 has the function of encoding members of the VEGFR family.These proteins play in angiogenesis,as well as aspects of the recasting of the role is crucial.According to the results and data we have currently studied,LncRNA plays an important role in the growth and development of the body and the development of the disease.However,in pre-eclampsia,especially in severe preeclampsia placenta tissue which,at present our study of LncRNA,its basic structure,distribution,changes in the activity of specific biological function,mechanism of action and future development yet Thorough and clear.Objective:Discussion of long chain non-coding RNA(long non-coding RNA,lncRNA)-LOC391533 involved in severe preeclampsia placenta correlation spiral arteries recast insufficient.Materials and Methods:1.Research object and inclusion criteria:The selection time is from January to December 2016,and the selected place is the Third Affiliated Hospital of Zhengzhou University.Select the object severe preeclampsia group: 36 cases of pregnant women diagnosed with severe preeclampsia clinically.The control group: According to the proportion of the selected period of the same period is 1: 1,pregnancy within weeks of gestational age difference,the age difference within 2 weeks,the normal prenatal examination of 36 healthy pregnant women.Inclusion criteria: Two groups of pregnant women,Chinese,and no labor,all were singleton pregnancies,no other pregnancy complications and complications,and no other chronic medical history.2.experimental method:2.1 The Western-blot experimental method was used to detect the protein levels of VEGF and SVEGFR-1 in placenta of preeclampsia and normal pregnancy groups,respectively.2.2 ELISA was used to detect the protein levels of VEGF and SVEGFR-1 in maternal serum in the preeclampsia group and the normal pregnancy group.2.3 The use of optical microscopy combined with scanning electron microscopy measurements of preeclampsia,normal pregnancy group,placental tissue spiral arterial wall thickness,lumen size were detected.2.4 RT-PCR using the experimental method of preeclampsia,respectively,the content of the two sets of normal pregnancy placenta LNCRNA-LOC391533 mRNA detected.3.Statistical analysis:We use SPSS 21.0 related system software to perform statistical analysis on the experimental data.X ±s is used for the measurement data and data that are in accordance with the normal distribution.Two independent samples t test are used for comparison between the preeclampsia group and the normal pregnancy group.;for eligible and non-normal distribution of data using the measurement data M(min ~ max)represents between preeclampsia and normal pregnancy group two groups were compared using Mann-Whitney U test.Between each group,the correlation between the data was processed using the Spearman correlation analysis method.For each data processing,P<0.05 was considered statistically significant.Results1 The LNCRNA-LOC391533 mRNA content in the placenta of severe eclampsia patients(2.45 ± 0.527)was significantly higher than that in the normal pregnancy group(1.0 ± 0.2),and the difference was statistically significant(P < 0.05).2 severe preeclampsia placental spiral artery wall thickness(122.6 ± 9.5)?m,compared with the normal pregnancy group placental spiral artery wall thickness(98.9 ± 2.5)?m thick,the difference was statistically significant(P <0.05)3 severe preeclampsia placental spiral artery lumen area(130.1 ± 22.3)?m2,compared with the normal pregnancy group placental spiral artery lumen area(188.1 ± 21.5)?m2 small,the difference was statistically significant(P <0.05)4 Protein levels of VEGF(0.2 ± 0.0),(28.1 ± 3.2)in placenta and maternal plasma in severe eclampsia were significantly higher than those in placental tissue and maternal serum in normal pregnant women(0.6 ± 0.1),(40.8 ± 3.2)The protein content was small,the difference was statistically significant(P<0.05)5 Protein levels of SVEGFR-1(0.4±0.1),(50.8±6.1)in placenta and maternal serum in severe eclampsia patients were significantly higher than those in placental tissue and maternal serum in normal pregnant women(0.2±0.0)The protein content of(15.6±2.4)was larger,the difference was statistically significant(P<0.05).6 The expression level of LNCRNA-LOC391533 mRNA in the placenta of patients with severe eclampsia was negatively correlated with the size of the lumen of the placenta spiral arteries(r=-0.702),and the degree of wall thickness was positively correlated(r=0.683).7 Severe eclampsia placenta expression levels LNCRNA ~ LOC391533 mRNA content size are positively correlated with the level of expression of placental and maternal serum sVEGFR-1 protein into the content size(r = 0.759)(r = 0.857),and maternal placenta and There was a negative correlation between the expression levels of VEGF protein in serum(r=-0.806)(r=-0.796)Conclusion:LNCRNA-LOC391533 RNA may be involved in the formation of severe preeclampsia by affecting the underrecirculation of placenta spiral arteries.
