Hyperlipidemia is closely related to the death rate of various cardiovascular diseases.Early treatment of hyperlipidemia can effectively control the incidence and mortality of cardiovascular diseases.However,the fact is that people generally do not have enough knowledge about the prevention and treatment of hyperlipidemia.Many patients with hyperlipidemia are not sufficiently active in the treatment of hyperlipidemia.Statins and fibrates,which are commonly used in clinical treatment of hyperlipidemia,can cause side effects such as liver damage.This is also an important reason for the lack of prompt treatment of hyperlipidemia.The Jindan Tablet studied in this article is composed of Rosa roxburghii,Hawthorn,Polygonum multiflorum,Peanut leaf,and Salvia miltiorrhiza.Among them,Peanut leaf and Hawthorn are dual-use medicinal herbs,which greatly reduces the toxic side effects during treatment.After pharmacodynamic evaluation of the pharmacodynamics and its efficacy in reducing blood lipids,we hope to conduct further research by finding out an analytical method suitable for compound Chinese medicine.Metabolomics,as a method of analysis in the field of compound Chinese medicine research in recent years,provides a new idea and new method for the study of traditional Chinese medicine and compound Chinese medicine.The research mainly consists of two parts:The first part is the pharmacodynamics evaluation of Jindan Tablet on hyperlipidemia model rats in order to determine the adjuvant effects of Jindan Tablet,a new compound preparation,on hyperlipidemia.Hyperlipidemia rat model was duplicated by intragastric administration of high-fat emulsion.The index components in the serum of rats,Total Cholesterol,Triglyceride,Low-Density Lipoprotein,and High-Density Lipoprotein were measured at different time points.to make sure that the model duplication is successful and that Jindan tablet has supportive therapeutic efficacy on hyperlipidemia rats.Meanwhile,body weight examination and livers morphology observation also showed that the Jindan tablet has a certain role in liver protection.In the second part,the pharmacological mechanism was further explored by means of the well-received metabolomics analysis method in traditional Chinese medicine pharmacological mechanism research,which greatly shortened the researchtime for the pharmacological mechanism of the compound Chinese medicine preparation.Metabolomic studies of rat serum were performed by Ultra-high Performance Liquid Chromatography-mass spectrometry,using Principal Component Analysis,Orthotopic Partial Least Squares-Discriminant Analysis with supervised patterns and software analysis.The metabolomics data were processed to obtain 88 and 137 different compounds and 22 and 37 related pathways,respectively.The trend of the differences in P(27)0.05 and the differences in lipid metabolism between these pathways were analyzed.The therapeutic mechanism of Jindan Tablet on hyperlipidemia is related to the regulation of Lecithin-Cholesterol Acyltransferase,and the protective effect on liver is related to Bile Acid-COA:Amino acid N-acyl-transferase.
