Objective Lipid abnormalities are closely related to the severity of SLE disease and the occurrence of various complications.In order to clarify the mechanism of abnormal lipid metabolism in SLE and explore the regulatory mechanism of Langchuangding Decoction.Multi-dimensional mass spectrometry "shotgun" lipidomics(MDMS-SL)technology were used to analyze the changes of lipid metabolism in liver tissue of MRL/lpr mice.Methods In this study,female MRL/lpr lupus mice and C57BL/6 mice as controls were used.The MRL/lpr lupus mice were randomly divided into four groups: model group,western medicine group,chinese and western medicine group,and chinese medicine group.Animals were fed continuously for 10 days after being fed adaptively.After sacrifice,the same position of the middle part of the largest leaf of the liver was harvested,and then the tissue homogenate was prepared after ultrasonication.Lipid extracts were prepared using a chloroform/methanol solvent system and analyzed by the MDMS-SL technology to find the differences in lipid molecules of each group.Results The MDMS-SL technology was used to qualitatively and quantitatively analyze 10 groups of lipid molecules including Car,TAG,PI,PC,PE,LPE,SM,LPC,PG,and HNE in each group of liver tissues.Compared with the model group,the content of Car and HNE in the model group increased significantly,and the increase range were about 54% and ~37%,respectively.The content of TAG was significantly reduced,reaching ~31%.Different types of LPC molecules appeared differently.The trend of change: sn-1 position LPC has an increasing trend,sn-2 position LPC content has a decreasing trend,PI,PC,PE,LPE,SM,PG and other lipid content have no significant changes.After drug intervention,the total amount of Car was recovered,the largest in the Chinese medicine group,followed by the Western medicine group,and the smallest in the Chinese and Western medicine group.The decrease in total HNE content was greatest in the Chinese medicine group,followed by the western medicine group,and the lowest in the Chinese and Western medicine groups.The largest increase in the total content of TAG was in the chinese medicine-treated group,followed by the western medicine group,and the lowest in the Chinese and Western medicine groups.Conclusions 1)Mitochondrial Energy Metabolism dysfunction in liver of MRL/lpr Lupus Rats;2)The activity of phospholipase PLA2 in liver tissue of MRL/lpr lupus rats is increased,which is closely related to the enhancement of SLE inflammation.3)All kinds of abnormal lipids in liver tissue of rats with lupus erythematosus showed a normal trend after intervention of Chinese medicine compound,which indicated that Langchuangding Decoction can effectively regulate abnormal lipid metabolism and improve the pathological state of SLE.
