Objective: This study is aimed to investigate the possible role of mi R-223 in the pathogenesis of primary gout inflammation.Methods:(1)The peripheral blood of 65 acute gout patients(AG group),45 interval gout patients(IG group)and 45 healthy controls(HC group)were collected.(2)The RAW264.7 mouse macrophage was stimulated with 200?g/ml MSU crystal for 3h,6h,and 12 h.Then the RAW264.7 culture medium and cells were collected to detect the relative level of NLRP3 mRNA and IL-1? protein.(3)The RAW264.7 mouse macrophage was transfected with miR-223 mimics,mi R-223 mimic negative control,miR-223 inhibitor,and miR-223 inhibitor negative control,the relative level of NLRP3 mRNA,and their protein,IL-1? protein of all group cells were detected.(4)mi R-223 was detected using Taq-Man PCR,the relative level of NLRP3 was detected using Real time fluorescence quantitative method.IL-1? protein was detected using Enzyme linked immunosorbent assay.NLRP3 protein was using Western Blot.(5)The measurement data were compared by one factor analysis of variance test.SPSS21.0 statistical software was used for analysis.Results:(1)The expression of miR-223 in AG group was lower than that in IG group(p<0.05).The expression of miR-223 in IG group was lower than that in HC group(p<0.05).(2)The miR-223 expression was all lower than that in control group when RAW264.7 was stimulated with MSU crystal for 3h,6h,and 12h(p<0.05).The expression of NLRP3 mRNA was all higher than that in control group when RAW264.7 was stimulated with MSU crystal for 3h,6h,and 12h(p<0.05).The concentration of IL-1? was all higher than that in control group when RAW264.7 was stimulated with MSU crystal for 6h and 12h(p<0.05).Compare to the group of 0h,the miR-223 expression was the lowest level.Contrarily,the expression of NLRP3 mRNA was the highest level.(3)The relative level of NLRP3,NLRP3 protein,and IL-1? protein in mi R-223 overexpression group was lower than that in control group(p<0.05).The relative level of NLRP3 mRNA and their protein and IL-1? protein in miR-223 inhibitory pression group was higher than that in control group(p<0.05).Conclusion: The differential expression of miR-223 in patients with acute gouty arthritis,intermittent gouty arthritis,and normal people,suggesting that mi R-223 may be involved in the regulation of gout inflammation.At the same time,mi R-223 can promote the spontaneous remission of acute gout inflammation by negative controlling NLRP3.
