As a serious disease that seriously threatens human health,obesity has become an important public health issue that has caused global concern.More and more research shows that obesity is an important cause of high incidence of various diseases,such as type II diabetes,fatty liver,insulin resistance,cardiovascular disease,cancer and other diseases.Cordycepin is one of a variety of biologically active ingredients isolated from the valuable Chinese herbal medicine Cordyceps sinensis.Cordycepin has resistance to inflammatory reactions,cures viral diseases,regulates immunity,reduces blood sugar,lowers blood fat,treats tumors,resists pathogenic microorganisms,etc.Many aspects of pharmacological effects.However,whether the effective prevention of obesity by cordycepin and its related mechanisms has not been reported in the literature.In recent years,the elevation of prolactin(PRL)has been shown to be closely related to food absorption and weight gain,and a large number of studies have shown that obesity can cause disorders of the intestinal flora.Therefore,this study intends to investigate whether cordycepin can effectively prevent obesity and how to prevent obesity by inhibiting the secretion of prolactin and regulating the abundance of intestinal flora.This article has done research on the following three aspects,the main research contents and results are as follows: 1.Cordycepin prevents obesity induced by high fat diet in ratsHigh,medium,and low-dose cordycepins were used to intervene in high-fat diet-induced obese rats to determine the pathological changes in body weight,body fat index,related biochemical parameters,adipose tissue,and liver tissue after 5 weeks of drug intervention.The experimental results showed that high,medium,and low doses of cordycepin significantly inhibited the increase in body weight,decreased the fat coefficient,and lowered serum cholesterol(TC),triglyceride(TG),low-density lipoprotein(LDL),and malondialdehyde aldehyde(MDA),aspartate aminotransferase(AST),alanine aminotransferase(ALT)and liver TC and TG contents(P < 0.01),and significantly increase the activity of superoxide dismutase(SOD)(P <0.01).However,high-density lipoprotein(HDL)only significantly increased in the high-dose group(P < 0.05).The results of liver pathology showed that high,medium,and low doses of cordycepin all had protective effects on liver damage induced by high-fat diet.The results of pathological section of fat showed that high,medium and low doses of cordycepin could significantly inhibit the increase of adipocyte.2.The mechanism of cordycepin inhibiting the secretion of PRL(1)The ELISA method was used to detect PRL in serum and the expression of PRLR and Jak2 in fat was detected by Western Blot.The results of in vivo experiments showed that the high,medium and low doses of cordycepin can significantly reduce the serum PRL content,and can significantly reduce the expression of PRLR in vivo,but have no significant effect on the expression of JAK2.(2)GH3 and 3T3-L1 cells were cultured in vitro and the cytotoxicity of cordycepin on cells was detected by CCK8 method.The expression of ADORA1,p-PI3 K,p-AKT,p-ERK,PRLR and JAK2 protein levels was detected by Western Blot.The effect of cordycepin on the secretion of prolactin was detected by ELISA and other methods.The differentiation of 3T3-L1 cells by prolactin was observed by fluorescence staining.The experimental results showed that cordycepin had no effect on the proliferation of GH3 cells when it was below 50 ?g/ml.Cordycepin could induce the expression levels of ADORA1,p-PI3 K and p-AKT in GH3 cells and inhibit the expression of p-ERK.High concentrations of prolactin can significantly promote the differentiation of 3T3-L1 cells into mature adipocytes and upregulate the expression of PRLR and JAK2 proteins.3.The effect of cordycepin on intestinal microflora in ratsHigh-throughput 16 S rDNA sequencing was performed on the intestinal contents of each group of rats after 5 weeks of experiments on the MiSeq Illumina platform.The results showed that cordycepin can significantly increase the diversity of the intestinal flora and reverse the long-term high-fat diet caused by the intestinal tract.The flora is disordered.In summary,the results of this study indicate that cordycepin in high-,medium-,and low-dose groups has good effects on preventing obesity and lowering blood lipids,and its mechanism of weight-loss and lipid-lowering may be through cordycepin on prolactin and intestinal bacteria.The adjustment of the group will provide new ideas for future research.
