Objective:To investigate the effect of electro-acupuncture on protecting weight gain caused by rosiglitazone administration in type ? diabetic(T2DM)rats,and its possible mechanism in CNS.Methods:SD rats with age of 3 weeks old were divided randomly into two groups:normal food(NF)group and diet induced obesity(DIO)group.After 4 weeks of dietary manipulation,DIO rats received an intraperitoneally injection of STZ to induce T2DM[non-fasting plasma glucose>16.67 mmol/1(300 mg/dl)].The rats in the NF group were divided into two groups:Control(Con)group and Control-electro-acupuncture(Control-EA)group.The T2DM rats were divided into four groups:T2DM group,T2DM-electro-acupuncture(T2DM-EA)group,Rosiglitazone(RSG)group,and Rosiglitazone-electro-acupuncture(RSG-EA)group.All rats including the control rats were under physical restraint.Rats in the RSG group and RSG-EA group received rosiglitazone orally at dose of 5mg/kg once a day.All rats in the EA groups were electro-acupunctured at the acupoints Zusanli(ST36)and Neiting(ST44).Daily food intake and weekly body weight change were monitored;insulin,triglyceride,cholesterol,and blood glucose levels were measured.Thalamus tissues were harvested at the end of experiments for further molecular analyses.Results:Body weight of these rats in RSG group were significantly higher than those in the T2DM group,but lower than those in the RSG-EA group.Increased food intake was observed in the RSG group.,Whereas,the rats in the RSG-EA group had reduced food intake and body weight compared to those in the RSG group.Furthermore,we detected that the expression of Ppar-g in thalamus is increased by gave rosiglitazone,while it reduced after EA.The expression of Leptin and STAT3 were increased as well in the RSG-EA group.HE staining indicated that the EA can narrow the shape of white adipose cells in RSG group.Conclusion:Electro acupuncture is an effective approach for inhibiting weight-gain in T2DM rats when they were treated by rosiglitazone.The possible mechanism might be through increased Leptin and STAT3,and decreased Ppar-g expressions,which may contribute toreduced food intake and weight-gain.
