| ObjectiveIn order to provide experimental basis for therapy that Schisandra lignin cure osteoporosis,the therapeutic effect of Schisandra lignin on osteoporosis ofovariectomized rats was observed,and the function of Schisandra lignin could treat osteoporosis was partly bright to light from the angle IL-6 and IL-11 which play an important role in regulating bone metabolism.Materials and Methods92 femal SD rats,SPF grade,Weighing 210±20g,were randomly divided into three groups,including a blank group(n= 12),a sham-operated group(n= 12),and a modeling group(n=68).The rats in modeling group were anesthetized with sodium pentobarbital 45 mg/kg and bilateral ovaries removed.For sham-operated group,only a little fatty tissue around bilateral ovaries was removed,one rat died in the procee operation.three weeks later after the operation,sixty-seven rats in the modeling group were divided again into 5 groups,a model group(n= 12),a positive control group(n=14),small-dose Schiandra lignin group(n= 13),medium-dose Schiandra lignin group(n=14),large-dose Schiandra lignin group(n=14).Then the small-dose,medium-dose and large-dose groups were fed with the medicine of Schiandra lignin for rats:8.05mg/ml,16.10mg/ml,32.20 mg/ml respectly;the dose volume is 12ml/kg;The dose of Schiandra lignin for rats:small-dose group:96.6 mg/kg,medium-dose group:193.2 mg/kg,large-dose group:386.4 mg/kg.The progynova in positive control group was 0.015mg/ml,the volume was 12ml/kg,and the dose was 0.18mg/kg;The drugs were given once a day and last six days,followed by one day of rest,and for the next six days,The whole period lasted thirteen weeks.The rats would be weight once a week,and the dose of the drug would be given according to the weight.Then the rats in blank,sham-operated and model group were given 1.07%twain solution with the same volume.After the last adminstration,The rats were anesthetized with continal(45mg/kg),After being killed,Detection of bone tissue morphology index would be carried out by micro-CT with their left femoral bone.their right tibia were made into frozen sections to detect the expression of IL-6,IL-11 albumen and mRNA in intibia bone marrow.The experimental datas were expressed by mean ± standard deviation(x±s),and analyzed by means of SPSS20.0 software.If the data were normally distributed,single factor analysis of variance were used for processing,if the homogeneity of variance,between the two groups were compared with LSD test,or Dunnett’s T3 methods were adopted.If the results were non normal distribution datas,Kruskal-Wallis 1-way ANOVA(k samples)test were adopted,and for the comparison between the two two groups,Stepwise step-down method was to be used.Results1 Influences of Schiandra lignin on target of bone histomorphometry of OVX rats1.1 Influences of Schiandra lignin on femoral bone BV/TV in OVX ratsIn contrast to sham-operated group,The rats’ femoral bone BV/TV in model,positive control and all the Schiandra lignin groups all reduced obviously.Compared with model group,The rats’ femoral bone BV/TV in medium-dose,large-dose and positive control groups all increased obviously,While the small-dose group showed no obvious difference.In contrast to small-dose group,The rats’ femoral bone BV/TV in medium-dose and large-dose increased outstandingly,While there was no clear difference between the two groups.1.2 Influences of Schiandra lignin on femoral bone Tb.N in OVX ratsCompared with sham-operated group,The rats’ femoral bone Tb.N in model,positive control and all the Schiandra lignin groups all decreaseded significantly.In contrast to model group,The rats’ femoral bone Tb.N in medium,large-dose and positive control groups all increased clearly,While the small group showed no obvious difference.Compared to small-dose group,The rats’ femoral bone Tb.N in medium and large-dose increasedly distinguishedly,While there was no distinct difference between the two groups.1.3 Influences of Schiandra lignin on femoral bone Tb.Th in OVX ratsIn contrast to sham-operated group,The rats’ femoral bone Tb.Th in model,positive control and all the Schiandra lignin groups all reduced obviously.Compared with model group,The rats’ femoral bone Tb.Th in medium,large-dose and positive control groups all increased obviously,While the small group showed no obvious difference.In contrast to small-dose group,The rats’ femoral bone Tb.Th in medium and large-dose increased conspicuously,While there was no clear difference between the two groups.1.4 Influences of Schiandra lignin on femoral bone Tb.SP in OVX ratsIn contrast to sham-operated group,The rats’ femoral bone Tb.SP in model,positive control and all the Schiandra lignin groups all increaseded significantly.Compared with model group,The rats’ femoral bone Tb.SP in medium,large-dose and positive control groups all decreased clearly,While the small group showed no obvious difference.Compared to small-dose group,The rats’ femoral bone Tb.SP in medium and large-dose decreased outstandingly,While there was no distinct difference between the two groups.1.5 Influences of Schiandra lignin on femoral bone SMI in OVX ratsIn contrast to sham-operated group,The rats’ femoral bone SMI in model,positive control and all the Schiandra lignin groups all increaseded significantly.In contrast to model group,The rats’ femoral bone SMI in medium,large-dose and positive control groups all decreased clearly,While the small group showed no obvious difference.Compared to small-dose group,The rats’ femoral bone SMI in medium and large-dose decreased extraordinarily,While there was no distinct difference between the two groups.2 Influences of Schiandra lignin on deliverances of IL-6 albumen and mRNA in OVX rats’ tibia bone marrow.In contrast to sham-operated group,The deliverances of IL-6 albumen and mRNA IOD in OVX rats’ tibia bone marrow in model and the small-dose groups all increased obviously.The deliverances of IL-6 albumen IOD which in medium,large-dose and positive control groups had no significant differences.While the IOD of IL-6 mRNA in medium-dose increased distinguishedly,but that in large-dose and positive control groups had no outstanding diversity.Compared with model group,The deliverances of IL-6 albumen and mRNA IOD in medium,large-dose and positive control groups decreased clearly.But those in small-dose group expressed no obvious differences.In contrast to small-dose group,The deliverances of IL-6 albumen and mRNA IOD in medium and large-dose group decreased obviously,while there were no clear difference between large-dose and medium groups.3 Influences of Schiandra lignin on deliverances of IL-11 albumen and mRNA in OVX rats’ tibia bone marrow.In contrast to sham-operated group,The rats’ deliverances of IL-11 albumen and mRNA IOD in OVX rats’ tibia bone marrow in model,small-dose and medium-dose groups increased obviously,The deliverances of IL-11 albumen IOD in large-dose group increased conspicuously,but the deliverances of IL-11 mRNA IOD in which had no outstanding divergences.while those in positive control group revealed no distinguished divergences.Compared with model group,The rats’ deliverances of IL-11 albumen and mRNA IOD in OVX rats’ tibia bone marrow in medium,large-dose and positive control groups all decreased obviously,While the smal-dose group showed no outsanding diversity.In contrast to small-dose group,The rats’ deliverances of IL-11 albumen and mRNA IOD in OVX rats’ tibia bone marrow in medium and large-dose descended conspicuously,While there was no distinguished diversity between the two groups.Conclusions1.BV/TV,Tb.Th and Tb.N in OVX rats’ femoral bone decreasedly clearly,but Tb.SP and SMI in those increased obviously,while Schiandra lignin could deteriorate the whole targets to a certain degree.It indicated that Schiandra lignin was useful to treating osteoporosis of rats which lost ovaries.2.Schiandra lignin can weaken the deliverances of enhanced IL-6’s and IL-11’s albumen in the rats’ marrow which leaded by losing ovaries,revealing that the drug could restrain the synthesis of IL-6 and IL-11 in the bone marrow and secretion to healing osteoporosis. |
