The purpose of this experiment is to investigate the effects of different addition forms and amounts of zinc and manganese on the quality of the bull's semen,and to screen the best amount of addition.Forty holstein bulls from the bull breeding station of Xi 'an Cow breeding Center were selected as experimental subjects and randomly divided into 10 groups with 4 bulls in each group and 4 replicates in each group.Control group and test group were set up.The control group was fed a basic diet + inorganic zinc sulfate(60mg/kg)and manganese sulfate(55mg/kg).The experimental group was fed a 3×3 experimental design with a basic diet + different levels of zinc methionine + different levels of manganese methionine.The amount of zinc methionine(in terms of Zn)is 60,120 and 180mg/kg,and the amount of manganese methionine(in terms of Mn)is 55,105 and 155mg/kg,respectively.Each group was fed continuously for 6 months,and semen was collected normally for 4 months.The results showed that the inorganic zinc-manganese control group and the organic zinc-manganese test group at the same level showed that the fresh sperm vitality,sperm density and production dose of the test group were significantly higher than the control group(P<0.05),and the sperm deformity rate of the test group was significant Lower than the control group(P<0.05);the number of sperm,acrosome integrity rate,and sperm motility of the bulls in the test group after thawing were significantly higher than the control group(P<0.05);in terms of sex hormone levels of the bulls,the test The levels of serum testosterone and androstenedione in the group were significantly higher than those in the control group(P<0.05),and the levels of serum estradiol in the test group were significantly lower than those in the control group(P<0.05).Comparing the test groups with different levels of organic zinc and manganese,it was found that the p H of fresh essence in each group was not significant(P=0.326),the production dose of each group was not significant(P=0.050),and the freshness precision of each group was significantly different(P=0.024),the difference in sperm motility of each group was not significant(P=0.107),the difference in sperm deformity rate of each group was not significant(P=0.140),the difference in the amount of sperm collection was not significant(P=0.215),but each group Comparison of comprehensive index of fresh sperm between 120mg/kg zinc methionine + 155mg/kg manganese methionine group,the highest sperm collection,sperm motility and lowest sperm deformity rate,the serum testosterone levels of each group were not significantly different(P=0.333),serum estrogen 2 Alcohol levels were not significantly different(P=0.357),androstenedione levels were not significantly different(P=0.070),the serum testosterone and androstenedione levels were highest in the 120 mg/kg zinc methionine +105 mg/kg manganese methionine group,and the serum female The level of diol was the lowest,and there was a significant difference in the number of straight sperm after thawing(P=0.000),there was a significant difference in the acrosome integrity rate after thawing(P=0.038),and there was a significant difference in sperm motility after thawing(P=0.043),the effective sperm dose after thawing is significantly different(P=0.015),in which zinc methionine 120mg/kg + manganese methionine 105mg/kg group thawed straight sperm number,acrosomal integrity rate,sperm motility and effective dose are the highest levels after thawing.Through comprehensive analysis and comparison,it can be seen that in the experimental group,fresh sperm vitality,fresh sperm density,fresh sperm production dose,sperm count,sperm intact rate,sperm vitality and effective semen dose after linear thawing,serum testosterone and androstenedione The level is better than the control group.In the test group,the above-mentioned levels of the zinc methionine 120mg/kg + manganese methionine 105mg/kg group are better than other groups.This group can be used as the preferred reference dose for improving the quality of semen before and after thawing of bulls.
