Epidermal growth factor(Epidermal growth factor,EGF)is an important factor in the development of goat mammary glands.As an important growth factor,it participates in the proliferation and development of mammary gland cells.In this study,the primary breast epithelial cells of dairy goats in the prime of lactation were selected as the test materials,and the phosphorylation level of EGF-mediated signaling pathway protein in goat mammary epithelial cells was detected.Synchronization was used to obtain the mitotic phases of goat mammary epithelial cells.Cells,and studied the effect of EGF on lipid metabolism of goat mammary epithelial cells in G2/M stage,and the results obtained are as follows:This study detected the phosphorylation level of EGF-mediated signaling pathway protein in goat mammary epithelial cells,and found that the phosphorylation of EGFR in mammary epithelial cells was activated,and the downstream PI3K/AKT and MEK/ERK pathways were also activated,specifically Inhibitors can down-regulate the phosphorylation of the above signal proteins.After obtaining breast epithelial cells of different phases through the synchronization scheme,the difference of triglyceride and cholesterol was detected.The results showed a significant increase in triglyceride and cholesterol content in G2/M breast epithelial cells(P<0.01).It was also found that EGF increased the triglyceride content in G2/M breast epithelial cells.The results of using specific AKT and ERK pathway inhibitors indicate that inhibition of AKT and ERK pathways can significantly reduce triglyceride content.In order to further verify the effect of EGF on G2/M breast epithelial cells milk fat metabolism related genes.This study detected triglyceride synthesis related genes ATPAT6 and DGAT genes,lipid droplet synthesis related genes XDH and TIP47 genes,triglyceride transport related genes FABP3 and FABP4 genes,and fatty acid synthesis related genes PPARG,SREBP1,FASN and SCD1 genes.The ACOX1,TAGL,and CPT1 A genes related to triglyceride breakdown were also detected.The results showed that EGF up-regulated the expression of DGAT,TIP47,ACOX1,TAGL and CPT1 A genes,and decreased the expression of SREBP1,FASN,SCD1,XDH and FABP3 genes.To sum up,this study found that EGF can activate EGFR-mediated signaling pathways in goat mammary epithelial cells,and the downstream PI3K/AKT and MEK/ERK pathways are activated accordingly;signaling protein inhibitors can block EGF-mediated signaling pathways.Signal protein activity.Cell synchronization reagents can significantly arrest goat mammary epithelial cells in each cell phase.EGF can up-regulate the content of triglycerides and lipid droplets in goat mammary epithelial cells in G2/M phase,mainly by up-regulating the DGAT gene and TIP47 gene expression to achieve. |
