Alternative splicing(AS)is an important mechanism for higher organisms to regulate gene expression and generate proteome diversity.It is a key influencing factor for higher organisms to produce diverse proteins and complicate transcripts.Signal transduction,biological and abiotic stress and other processes all have important regulatory effects.Selective shearing is widely involved in the transcriptional regulation of higher organisms against biological stress or abiotic stress,and provides organisms with a timely and effective regulation scheme that responds quickly to environmental changes.This study is based on a large number of transcriptome and genomic data of different Bemisia tabaci cryptic species that have been sequenced in the database.First,the transcriptome data is processed and analyzed for quality control.The HISAT2 and Bowtie2 software are used for comparative analysis of sequence conditions.Hidden selective shearing differences between species,quantifying the expression of variable shearing events,and finally performing GO analysis and KEGG Pathway enrichment analysis on differentially expressed genes.The following research results were obtained:1.The identification and comparison analysis of the selective shearing of S and C groups of the three Bemisia tabaci cultivars showed that the selectivity ratios of the three types of Bemisia tabaci,MEAM1,MED and SSA-ECA,were 2.3% and 0.9 %,1.3%.The proportions of variable shear(TSS)in the transcription start region and variable shear(TTS)in the transcription end region are the largest.In the MEAM1 subspecies,some genes related to calcium channels,ceramide metabolism,and ion-gated pathways were cut;in the MED subspecies,some genes related to ion-gated pathways and DNA binding sites were cut.;In the SSA-ECA pathway,some related genes such as ion-gated pathways and hormone level regulation pathways are sheared.2.Comparative analysis of MED type Bemisia tabaci at different developmental stages,adult and egg selective shearing,the result showed that the ratio of selective shearing was 1.9%,variable shearing at the beginning of transcription(TSS)and variable shearing at the end of transcription Cut(TTS)accounts for the largest proportion,mainly due to some genes related to biological protein,acetamide synthesis and other related pathways.3.The effect of selective shearing of MED Bemisia tabaci on virus infection,compared with TYLCV and ToCV,the result is that the proportion of selective shearing is 0.6%.Regional variable shear(TTS)accounts for the largest proportion,mainly due to some genes in protein synthesis and transfer,protein metabolism and other related pathways.4.Compared with the control,the Bemisia tabaci MEAM1 cryptic species Mo Shilang and the control showed that the proportion of selective shearing was 1.6%,and the variable starting shearing region(TSS)and variable ending shearing region(TTS)accounted for The largest proportion is mainly due to genes related to hormone regulation pathways,protein synthesis and other related pathways.By analyzing the difference of selective shearing of the Bemisia tabaci under different hidden species,different environmental stresses and biological stress conditions,the mechanism of adaptability to adversity and drug resistance was explored.From the perspective of selective shearing,it lays the foundation for excavating the damage mechanism of Bemisia tabaci invasion.
