Tomato(Solanum lycopersicum)is native to South America and has now become one of the most important crops in the world.At the same time,due to its small genome and easy genetic manipulation,tomato is a model research crop,and its position in agricultural and plant science research is increasingly prominent.Mutants are generated by EMS mutagenesis,and then the related mutant genes are screened to study the function of genes,which has become an important aspect of functional genomics research.In this paper,the dwarf mutant obtained by sequencing tomato material 'Heinz 1706' EMS mutagenesis was used as the main test material,the characteristics of the mutant at different developmental stages were analyzed,and the shoot tip material was taken for transcriptome analysis to try to screen for possible mutation-related Gene,the main results of the paper are as follows:1.It is clear that the IAA dwarf mutant of tomato'Heinz 1706' EMS dwarf mutant has a greater effect on hypocotyl elongation,while GA3,which has a greater effect on tomato stem elongation at seedling stage,has a greater effect on tomato fruit development it is also GA3.2.Transcriptome sequencing was performed on tomato stem tip mixed samples.A total of149.02 Gb high quality sequence bases were obtained by sequencing.The number of high quality sequences in each sample reached 7.17 Gb,and the percentage of Q30 bases was91.97% or more.According to the statistical results of the comparison,the comparison efficiency between the Reads and reference genomes of each sample ranged from 82.00% to89.79%.Using the stem tip samples of tomato 'Heinz 1706' and its EMS dwarf mutants for sequencing,28,527,071 reads and 26,078,626 reads were obtained.The number of differentially expressed genes was 534,and the number of up-regulated genes was 426.The number of genes expressed is 108.3.Among the differentially expressed genes,four genes related to dwarfing hormones(auxin and gibberellin)were identified.Among them,Solyc03g006360.2and Solyc06g008580.2are related to auxin inhibitory protein and auxin-induced expression,while Solyc04g017720.2and Solyc02g083880.2 are related to gibberellin regulatory protein.Among them,the up-regulated genes are Solyc03g006360.2,Solyc06g008580.2and Solyc04g017720.2,and the down-regulated genes are Solyc02g083880.2.The four differential genes found were verified by fluorescence quantitative PCR,and the results were consistent with the results of transcriptome sequencing.It is further proved that these four differentially expressed genes may be involved in the dwarfing process of tomato ‘Heinz 1706' EMS dwarf mutant.
