| After this study using PCR amplification mitochondrial genome by PCR product direct sequencing,and analyzes the rice grasshopper four species of two genus mitochondrial genome,and obtained the sequence of its internal characteristics and organization structure,which is covered by controlled area and gene sequence and base composition,etc,at the same time also contains amino acid and the concrete conditions of the codon.Analysis of evolutionary characteristics and their phylogenetic relationship.The results are as follows:1.The mitochondrial genomes of Oxya hyla intricata 、 Oxya chinensis 、Pseudoxya diminuta amd Oxya japonica are all presented with the structure of double chain molecules.Meanwhile,the total length of its sequence is found to be 15 174 bp,15 486 bp,15 429 bp and 15 089 bp.The composition and sequence of the genes are the same as that of the currently published species of pterophididae,which contain the same genes as other arthropods,and the number of genes reaches 37,in addition to the A+T rich area,the number of which is 1.This has a high homology with the mitochondrial genome now tested.2.All the protein-coding genes of the four species were typical ATN initiation codons;Except for ND5 itself,which belongs to incomplete termination codon T,all the remaining protein-coding genes belong to relatively complete termination codon TAG and TAA.3.Based on 13 proteincoding genes and 2 r RNA genes of 64 species,Velarifictorus hemelytrus、Gryllotalpa unispina、Tettigonia chinensis and Gastrimargus marmoratus were selected as outgroups,and then the phylogenetic tree was constructed by maximum likelihood method(ML).The results showed that the main body of the cluster tree was divided into three clusters,and Oxya was the third largest cluster,which was located at the root of the cluster tree and had a close genetic distance with the outer group.4.The secondary structure of the four species of t RNA is veryconservative.Both amino acid and anticodon ring and TΨC ring,etc.,will produce different extent base mismatch mutation.5.A phylogenetic tree was constructed using the combined data set of amino acid sequences of 13 protein-coding genes in the mitochondrial genome.The main method used in this study and this study is the ML analysis method.Based on this method,the topological structure was obtained.This paper provides a reference for the internal sequence and architecture evolution of the mitochondrial genome in insects and orthoptera.This paper has a certain reference role for the evolution of the internal sequence and architecture of the insect and orthoptera mitochondrial genomes. |
