In this study,the original bulbs of Cremastra appendiculata were used as test materials to explore the influencing factors and induction pathways of cluster buds,Based on the successful establishment of the cluster bud induction method of C.appendiculata,the physiological and biochemical dynamic changes during the cluster bud development were explored..The main findings are as follows:1.The aseptic germination protocorms of C.appendiculata seeds were used as explant materials to explore the effects of different basic media,types of plant growth regulators,anti-browning agents and their concentrations on the induction of cluster buds of C.appendiculata.The results showed that:1/4 MS medium was the suitable basic medium for cluster buds growing;when the concentration of plant growth regulator TDZ was 2.0 mg·L-1 and the concentration of IAA was 0.2 mg·L-1,the induction rates of cluster bud were higher which are72.58%and 55.63%respectively;when the anti-browning agent was PVP and its concentration is 2000 mg·L-1,the induction rate of cluster buds was higher which is 79.90%.The suitable conditions for C.appendiculata cluster buds induction are:1/4 MS+TDZ 2.0 mg·L-1+IAA0.1 mg·L-1+PVP 2000 mg·L-1,and the induction rate can reach 81.16%.2.The content of soluble sugar and soluble protein changed regularly during the clustering of C.appendiculata.The soluble sugar content decreased at the beginning,and then increased under each treatment.The soluble sugar content decreased from the start of cluster bud induction to the initial induction period,then increased and maintained at a high level during the middle to late induction period.In the process of cluster bud development,the soluble protein content was generally"M"type.The soluble protein content increased from the start of cluster bud induction to the initial induction period,and the soluble protein content decreased during the early to middle induction period.The content of soluble sugar and the content of soluble protein are mainly negatively correlated.The content of soluble protein is significantly negatively correlated with the carbon-nitrogen ratio,and the induction coefficient is higher than that of other treatments.The change of the carbon-nitrogen ratio mainly showed a downward trend and then an upward trend.To sum up,the occurrence of azalea cluster buds is comprehensively regulated by the changes of soluble sugar and soluble protein content.3.The changes of endogenous hormones(abscisic acid ABA,zein ZT,3-indoleacetate IAA,gibberellin GA3)during C.appendiculata sprouting were measured,and the changes of the endogenous hormone ratio during the sprouting were analyzed.The results show that:the content of different endogenous hormones varies during the development of cluster buds;from the initiation of cluster buds to the middle stage of induction,the contents of IAA,GA3,and ZT increase,and the content of ABA decreases;during the induction period,the contents of ABA and IAA decrease As the content of GA3 and ZT decreased,the content of IAA decreased and the content of ABA,GA3and ZT increased.During the clustering process,the ABA/GA3 ratio showed a downward trend from the induction to the middle period,and it showed an upward trend during the induction period;the ZT/IAA ratio showed a downward trend from the induction to the middle period,and it showed an upward trend from the middle period to the later period.
