Endophytic fungi of medicinal plants are a kind of microbial resources rich in bioactive components.In this study,Scale-up fermentation the endophytic strain(Phomopsis sp.By254)of Polygonatum sibiricum was carried out and silica gel column chromatography,Sephadex LH-20 chromatography as well as reversed-phase Rp-C is column chromatography were used to separate and purify the secondary metabolites in ethyl acetate extraction of fermentation products.For the isolated compounds,their structures were identified by means of MS and NMR(1H-NMR,13C-NMR,H-HCOSY,HMBC,HMQC,NOESY).The final results were as follows(1)Fourteen compounds were isolated and purified from the crude extract of ethyl acetate phase,and identified as trichodermamide A(I),didymellamide B(II),phomoxanthone A(III),ticerandrol B(IV),19,20-epoxycytochalasin-Q(V),cytochalasin N(VI),kodo-cytochalasin-1(VII),cytochalasin-J(VIII),vitamin-B2(IX),(+)-scytalone(X),?-sitosterol(XI),methyl gallate(XII),gallic acid(XIII),4-hydroxy-2-methyl-3-oxocyclohepta-1,4,6-tnene-l-carboxylic acid(XIV)(2)Among them,compound XIV was discovered for the first time.In addition,compounds I,II,IX,X,XII and XIII were isolated from endophytic fungi of Polygonatum sibiricum for the first timeThe above study enriched the research content of P.sibiricum,and provided a reference for further exploration of the secondary metabolites of its endophytic fungi.
