Halenia elliptica D.Don belongs to Halenia genus in the Gentianaceae family, one of the commonly used medicines for the treatment of diseases of the hepatobiliary system in Tibetan medicine system. In this paper, based on UPLC/Q-TOF-MS/MS non-targeted metabolomics technology, the changes of Halenia elliptica D.Don metabolites and characteristics of metabolic pathways in different habitats and geographical flora were studied with multivariate statistical analysis method and a variety of Web working platforms and KEGG pathway analysis. The main results were as follows:1.According to the geographical structure of Qinghai Province,22 samples were divided into four groups: Datong River Basin in Qilian Mountains,Hehuang Valley,The Mother River Basin in Animaqing region and Make River Basin in Qingnan plateau in Hengduan Mountains.According to the characteristics of vegetation habitat and community structure, they were divided into four groups: meadow, shrub, forest and gravel.2.Based on UPLC/Q-TOF-MS/MS technology for wild Halenia elliptica D.Don metabolites in different regions of the study found that:(1)According to PCA analysis and hierarchical clustering heat map double verification, the samples of Make River Basin,Hehuang Valley,Datong River Basin and Animaqing the Mother River Basin are all in a single group.(2)All the different metabolites were concentrated in 49 metabolic pathways.Among the six groups, through the analysis of KEGG enrichment, the most significant pathway was galactose metabolism pathway and valine, leucine and isoleucine degradation pathway, among which valine, leucine and isoleucine degradation pathway might be related to drought environment stress, and galactose metabolism pathway could be related to root growth and development at high altitude.3.Based on UPLC/Q-TOF-MS/MS technology on Animaqing region of the Mather River basin under the condition of different habitats Halenia elliptica D.Don metabolites study found that:(1)According to PCA analysis, there is a single group ofgravel in grassland and river beach, and there is a group of gravel in shrub and under forest. The results of hierarchical cluster analysis are basically consistent with PCA results, but there are significant differences in metabolites in shrub and under forest samples.(2)The analysis of the differential metabolites in each group showed that all the differential metabolites were concentrated in 59 metabolic pathways.According to the analysis of KEGG enrichment, galactose metabolism pathway was significantly different among the six groups.Galactose metabolism may be related to the growth and development of root system at high altitude, and galactose metabolism pathway may be less affected by the habitat, with a high correlation with the altitude.
