Cotton pigment glands,also known as gossypol glands,are unique structures of the genus cotton and its adjacent plants.Gossypol is closely related to the number and size of pigment glands.This study chose a glandular cotton,a dominant without glands cotton and a stealth as experiment materials,researching on pigment glands to form related transcription factors in spatio-temporal expression of different glands,analyzing the function of these transcription factors to help parse cotton glands molecular mechanism of formation,so as to foster new low phenol cotton varieties with candidate genes and auxiliary selection markers.The main results are as follows:1.The fluorescence quantitative PCR results showed that the relative expression of GhA01G0267 gene in the tissues of the glan-containing cotton is higher than that of the dominant and recessive glan-free cotton at different growth stages.GhD07G2328 gene in cotton glands ‘Zhongmiansuo12' in different growth period of cotton by the relative expression quantity of tissue is obviously higher than that of recessive without glands cotton‘ Hanwu198',with a few glands in the vegetative growth stage of cotton of ‘Zhongmiansuo12' main part of roots and buds and young embryo significantly better than the relative expression of dominant gland wool cotton of ‘Zhongmiansuo12 dominant without glands'.The spatio-temporal expression patterns of GhA01G0267 and GhD07G2328 genes are also highly correlated with the temporal and spatial distribution of gland density,suggesting that these two genes are closely related to the formation of cotton pigment glands and may be involved in regulating gland development or gossypol synthesis.2.The length of GhA01G0267 gene is 1743 bp,the length of coding area is 960 bp,and the code is 319 aa.There is one NAM domain between 22 and 155 aa.The molecular weight of the protein is 36.3 KD and the theoretical pI is 8.84.The prediction of subcellular localization is likely to locate in the nucleus,and the predicted threedimensional structure indicates that the 17~190aa residue sequence highly matches the stress-induced transcription factor NAC1 conserved sequence.The phylogenetic tree construction showed that GhA01G0267 is most closely related to cotton plants and more closely related to dicotyledon plants than monocotyledon plants.The conserved motifs of GhA01G0267 homologous protein indicated that motifs 3,5,1,2 and 4 were highly conserved in different plants.3.The total length of GhD07G2328 gene is 1542 bp,the encoding region is 501 bp,and the encoding is 166 aa.There is one WRKY domain between 87~146 aa.The molecular weight of the protein is 19.3 KD and the theoretical pI is 9.54.The subcellular localization is likely to locate in the chloroplast and participate in plant photosynthesis.The results of 3d structure prediction showed that the 74-148 aa residue sequences were highly compatible with the NPR1 conserved sequence.The phylogenetic tree construction showed that GhD07G2328 is more closely related to cotton plants and other dicotyledon plants than to monocotyledon plants.The conserved motifs of GhD07G2328 homologous proteins indicated that motifs 4,1,5 and 2 were highly conserved in different plants.
