| Cotton Verticillium wilt is a soil-borne fungal disease caused by Verticillium dahliae,which seriously affects the sustainable development of cotton production out and inside of China.The method for controlling this disease is especially to breed disease-resistant varieties,but the lack of effective anti-pathogenic materials limits the breeding speed of cotton varieties resistant to Verticillium wilt.In order to solve a series of problems such as the mechanism of interaction between cotton and pathogenic bacteria,the pathogenic mechanism of pathogenic bacteria,and the pathogenic process of pathogenic bacteria,etc,it is one of the important tasks to identify cotton secondary metabolites and their role in cotton disease resistance.To this point,this research proposal focus on analysis of the secondary metabolite expression level and temporal and spatial dynamic changes in the process of interaction between pathogen and cotton plant on upland cotton line 90472-4,so as to identify the important secondary metabolites of cotton related to cotton Verticillium Wilt and determine the important biomarkers.However,these scientific data provide the scientific basis for further study on metabolic pathways of these metabolites and biomarkers against Verticillium wilt on cotton.The main findings are as follows:1.The UPLC-Triple TOF-MS/MS detection method of cotton secondary metabolites was established。18 cotton samples had been detected with this method and dstermined of 2466 ESI+and 3371 ESI-metabolites presented in root,1823 ESI+ and 3721 ESI-metabolites presented in stem,and 2084 ESI+ and 2588 ESI-metabolites presented in leaf.2.A UPLC-Triple TOF-MS/MS standard library of 16 cotton metabolite standard products,including organic acids,ketones,terpenes,phenols,sugars and aldehydes,was constructed.3.The data were processed by statistical methods and the differentias between different treatments of same metabolites were analyzed.to obtain important differential metabolites.3208 important differential metabolites were obtained.By searching the standard library,only small part of these differentials mathched Gossypol,Melibiose and Astragalin. |
