In the world,artificial fertilization technology is widely used in pig production,pig semen preservation technology as an important step in the process of artificial insemination has been paid more and more attention.pig semen is vulnerable to various adverse effects during preservation,resulting in excessive ROS,damage to sperm structure,which greatly affects the quality of sperm after preservation.hence,natural efficient and non-toxic antioxidants need to be added to the semen preservation diluent to protect sperm from ROS damage and improve the quality of the preserved sperm.duroc boar semen was used in this trial study.different concentrations of citi pinein,kaempferol,and root endothelin were added to the porcine sperm preservation diluent.then,the activity and activity of pig sperm,integrity of acrosome and plasma membrane,mitochondrial activity,superoxide dismutase,catalase and glutathione peroxidase were detected respectively.MDA levels were detected by lipid oxidation kit.Finally,the best sperm quality concentration group was selected for in vitro fertilization.The results are as follows:1.The normal temperature preservation process were significantly improved after adding a certain concentration of citipine to the diluted solution(P<0.05).If the concentration of citisong in the diluent is 1.6 m M,the improvement of pig sperm quality is the best.when a certain concentration of citipine was added to the low temperature preservation diluent of pig sperm,the semen retention time could be extended to 9 d,d 9,the sperm viability and plasma membrane and acrosome integrity were significantly improved(P<0.05).when the concentration of citisong in the diluent was 1.6 m M,the preservation effect of pig sperm was the best.the mitochondrial activity,plasma membrane and acrosome integrity,and activity and viability of pig sperm were significantly increased after thawing by adding 1.2 m M of citipine in the cryopreservation diluent of pig sperm,which significantly decreased the MDA content and increased the activity of SOD and GSH-Px enzymes(P<0.05),and did not significantly improve the activity of CAT enzymes.2.Preservation diluent played a protective role on pig sperm during normal temperature,and the survival rate of pig sperm and the integrity rate of plasma membrane and acrosome increased significantly during the preservation process(P<0.05).at the concentration of kaempferol of 100μM,the improvement effect of sperm quality in pigs was the best.the addition of certain concentration of kaempferol in the preservation diluent has a good protective effect on pig sperm during cryopreservation.when the preservation time of semen was extended to 9 d,d 9,the viability of sperm and the integrity of plasma membrane and acrosome were significantly improved(P<0.05).when the concentration of citison in the diluent was 100μM,the preservation effect of pig sperm was the best.80μM of kaempferol was added to the cryopreservation diluent of pig sperm.after thawing,the mitochondrial activity,viability and activity rate,plasma membrane and acrosome integrity rate of sperm were significantly increased,which significantly decreased the MDA content and increased the activity of SOD,CAT and GSH-Px enzymes(P<0.05).3.The addition of a certain concentration of radicellin to the diluted solution of pig sperm preservation at room temperature,the viability of sperm and the integrity of plasma membrane and acrosome were significantly improved during preservation(P<0.05).and when the concentration of root endothelin in the diluent was 80μM,the improvement effect on pig sperm quality was the best.The survival rate,plasma membrane integrity and acrosome integrity of pig sperm(P<0.05)were significantly improved when the semen retention time was extended to 9th d after adding a certain concentration of root endothelin to the low temperature preservation diluent of pig sperm.pig sperm preservation was the best when the root pitin degree in the diluent was 80μM.the addition of 60μM of root endothelin to the cryopreserved diluent of pig sperm,the mitochondrial activity,activity and activity rate,plasma membrane and acrosome integrity rate of sperm were significantly increased after thawing,which significantly decreased the MDA content and increased the activity of SOD,CAT and GSH-Px enzymes(P<0.05).4.In the cryopreservation dilutions of pig sperm,the addition of citi pinein,kaempferol and root picotin,respectively,can significantly increase the penetration rate,single sperm entry rate and total fertilization rate of pig sperm during in vitro fertilization after thawing(P<0.05). |
