As one of the most important food crops in the world,rice is playing the pivotal role for the sustainal development.Also as the staple food of Chinese,the rice production is related to the issue of food for hundreds of millions of people.The early-senescence of rice is an important limiting factor in determining of the yield in rice.Therefore,isolation and characterization of the senescence associated genes is of great significance for exploring the mechanisms related to senescence of rice and benefit for breeding high-yield and high-quality rice.Previously,the rice early leaf senescence gene ELS6 had been cloned by map-based cloning approach in our lab.To further explore the molecular mechanism of ELS6 in leaf senescence,several apporoaches,such as biochemistry,physiology and genetics had been applied.1)To further examine the effect of ELS6 on leaf senescence,we transformed ELS6-GFP into els6 background and evaluate the phenotypes of transgenic plants(OE).The els6 began to show the senescence phenotype from the booting stage until the death in the mature stage.The plant height,number of tillers and chlorophyll content of the OE were all improved compared with WT.2)Analysis of the relative expression level of SAGs showed that: some SAGs were increased in els6 and significantly reduced in OE.3)The deletion of the ELS6 resulted in a smaller grain size,which was significantly increased in OE plants.4)There are many promoter elements related to hormone response in the promoter of ELS6.ELS6 is expressed in young plant organs highly and low in aging tissues and organs.ELS6 is localized in the cell nucleus.5)We identified an interaction protein of ELS6 is RBX1.6)ELS6 keeps the canonical role in Arabidopsis and Rice.7)The deletion of ELS6 causes the increase of ubiquitination in plants,and ELS6 is involved in the process of ubiquitination in plants.In summary,our data suggested the interaction of ELS6/RBX1 is to initiate thedegredation process of early leaf senescence genes.
