Yupingfeng polysaccharides(YPF-P)is an effective part of the total polysaccharides of Yupingfeng powder.Astragalus polysaccharides(APS),Atractylodes polysaccharides and Sposknikovan,are the main material basis of the clinical efficacy of this prescription.YPF-P and APS have extensive biological activities such as enhancing immunity,anti-stress and anti-virus,which have been widely used in animal husbandry and veterinary fields.At present,poor stability,low bioavailability,easy degradation and weak cell penetration were the disadvantages of the most traditional Chinese medicine polysaccharide preparations,which affect the efficacy of traditional Chinese medicine polysaccharides.Liposome is a kind of drug with the advantages of slow release,targeted administration,improved stability,and reduced adverse drug reactions.In recent years,it has been widely studied and applied in traditional Chinese medicine preparation.The main research contents are as follows:(1)YPF-P and APS were extracted by water extraction and alcohol precipitation;AB-8 macroporous resin was purified;polysaccharide content was determined by phenol-sulfuric acid method.(2)The monosaccharide composition type and mole ratio of YPF-P and APS were determined by TLC and pre-column derivatization HPLC method.(3)YPF-P liposome and APS liposome were prepared by reverse evaporation method;the conditions were optimized by orthogonal test;the enveloping rate was determined by ultracentrifugation method.(4)The morphology,particle size,potential distribution and chemical structure of liposomes were determined by transmission electron microscope(TEM),nanoparticle analyzer and Fourier transform infrared spectroscopy(FTIR).(5)The drug release of liposomes were investigated in vitro.(6)The liposomes were administrated,then the proliferation of chicken spleen lymphocytes were determined by MTT after 48 h.(7)The stability of liposomes were determined under different storage conditions and simulatedgastrointestinal environment.The results showed that:(1)The extracted crude polysaccharides content of YPF-P and APS were above 60%,and the purified polysaccharides content were above 90%.(2)YPF-P was composed of eight monosaccharides,including D-fructose,D-mannose,D-ribose,Rhamnose,D-galacturonic acid,D-glucose,D-galactose and Arabinose;the mole ratio was 6.31 : 3.13 : 1 : 5.38 : 2.21 : 126.18 : 41.13 : 66.99.APS was composed of five monosaccharides,including D-fructose,D-mannose,D-glucose,D-galactose and Arabinose;the mole ratio was 8.39 : 1: 263.41 : 11.26 :8.32.(3)The optimal technology for preparation of YPF-P liposome and APS liposome by reverse evaporation was following;the ratio of lecithin to cholesterol was 8:1,the ratio of lecithin to drug was 4:1,the ultrasonic time was 90 s;the encapsulation rates of YPF-P liposome and APS liposome were 88.68 % and 92.87 %,respectively.(4)The average particle size of YPF-P liposome was 231.4 nm;the polydisperse index(PDI)was 0.317;the Zeta potential was-30.6 m V.The APS liposome was 232.1 nm,the polydisperse index(PDI)was 0.242;the Zeta potential was-32.8 m V.Both of them were elliptical or spherical in shape,and no new chemical bonds were formed in their chemical structures.(5)The cumulative release rates of YPF-P liposome and APS liposome were 73.45 % and 62.67 % after 24 h,respectively.(6)The chicken spleen lymphocytes were proliferated significantly by YPF-P liposome and APS liposome alone or together with Con A.The effect of YPF-P liposome was better than APS liposome.The effect of liposomes of polysaccharides was significantly higher than the uncoated polysaccharides.(7)YPF-P liposome was stored in refrigerated(3 ? 5?)and room temperature,after 60 d the leakage rate were4.65 % and 42.61 % respectively;the APS liposome was 4.43 % and 32.28 %.Under PBS solution with p H 6.8 ? 8.0,artificial intestinal fluid and distilled water,both of them were relatively stable;under acid condition and artificial gastric juice,the stability needed to be improved.The encapsulation rate of polysaccharides liposomes were over 80% by this method.It have good slow-release property and stability and should be stored underthe condition of(3 ? 5?).The stability was excellent under the condition of neutral and artificial intestinal juice,and that was improved under acid condition and artificial gastric juice.The innovative research were carried out by this traditional Chinese medicine polysaccharides liposomes in the field of veterinary medicine.It have guiding significance for improving the bioavailability,stability and therapeutic effect of polysaccharide preparation.
