| Nuclear Polyhydrosis Virus disease in Bombyx mori is a disease caused by the nuclear polyhedrosis virus,BmNPV,which is the most common and most severe infectious disease in the sericulture industry.The chemical disinfectants are commonly used to strengthen the strict disinfection during the silkworm breeding period to prevent the infection and spreading of the virus.Although some researchers have found that some chemicals have a certain inhibitory effect on the pathogenesis of bombyx mori after infection with BmNPV,but it mainly reflects the preventive effect rather than the therapeutic effect,so there is no specific medicine for the treatment of this disease.Therefore,the development of BmNPV treatment drugs has great significance for the safety of sericulture industry.This study is aimed at the Nuclear Polyhydrosis Virus disease of Bombyx mori,the following were studied:Using molecular docking and virtual screening of small molecule compound databases,combined with cell and silkworm experiments to determine the efficacy of the drug;verifying the efficacy of some commercial antiviral and antitumor drugs in the treatment of BmNPV;investigation of a nano-antibacterial material on the prevention and treatment effect of silkworm disease.The main findings are as follows:1.Based on the mainstream drug development program,targeting the GP64 protein necessary for nuclear polyhedrosis virus’ replication that is pathogen of Nuclear Polyhydrosis Virus disease of Bombyx mori.First use virtual modeling technology to obtain its three-dimensional structure,and then use molecular docking to screen the ZINC small molecule database.The obtained small molecule compound ZINC95401182 was tested for activity.When ZINC95401182 was used at a concentration of more than 1μg/m L,it could completely inhibit the replication of BmNPV.It has certain treatment effect.2.Through the screening of 75 kinds of commercial antiviral and antitumor drugs,the therapeutic effect on Nuclear Polyhydrosis Virus disease of Bombyx mori was verified.The results showed Fenofibrate,rgx-104,STO 609,SMER 728,PK11195 and BEXAROTENE had significant antiviral effects at concentrations of 200μM,10μM,20 μM,100 μM,50μM,and 25μM.3.In order to screen for new disinfectants,this study investigated the effect of a nano-antibacterial material on the prevention and cure of silkworm disease.After reasonablely design experiments,the results showed that the nano-antibacterial material had a certain inhibitory effect on Beauveria bassiana and Bacillus bombyseptieus.However,it has no significant antiviral effect on BmNPV and no significant antiviral effect on CPV. |
