| The Hyphantria cunea(Drury)is an invasive polyphagous pest in the forest,which had destroyed more than 600 plants.In recent years,the hazards and host range of H.cunea is widening,but study of mechanism of adaptation to different host secondary metabolites aspect is still scarce.We determined the influence of plant secondary metabolites of quercetin,DNJ and chlorogenic acid on growth of the larvae of H.cunea and inducing detoxifying enzyme response,which is combing transcriptome sequencing.The results are as follows:1 The mortality of 5 instar larvae which was treated by different contents of quercetin,chlorogenic acid and DNJ for 6d were determined.The results show that the mortality of three secondary metabolite has significant difference(F=38.415,P<0.05;F=48.634,P<0.05;F=16.662,P<0.05).The three secondary metabolites had good pesticidal activity.The deaths of highest content group reached 70.00%,67.50% and 52.50%.2 The nutritional effects of different content of three secondary metabolite were determined.The results show that quercetin inhibited the feeding behavior of larvae.The RCR of treated group was significantly lower than the control group,but the ECD was higher than the control group except the group of 0.250%(P <0.05).Chlorogenic acid and relatively high contents of DNJ reduced utilization of nutrients for larvae.The ECD significantly of treated group significantly lower than the control group(P <0.05),but the RCR higher than that.3 The detoxification activity of H.cunea were determined processed by different content of the three secondary metabolite.In terms of content,quercetin has no significant effect on the Car E activity,which is similar to the chlorogenic acid on CYP450 activity.However,quercetin,DNJ and chlorogenic acid had significant effects on the other detoxification enzyme of H.cunea(P <0.05).The detoxification activity of H.cunea were determined processed by 0.500% three secondary metabolite for different time.In terms of time,three secondary metabolites had no significant effect on the Car E activity,which is similar to the DNJ on UGT activity.However,quercetin,DNJ and chlorogenic acid had significant effects on the other detoxification enzyme of H.cunea(P <0.05).4 Effects of three secondary metabolites on the growth of H.cunea were determined by 0.500% three secondary metabolite.When the content was 0.500%,the quercetin,chlorogenic acid and DNJ could extend the duration of larvae(3th to 7th)about 20.75%、14.354%、26.92%.Moreover,the developmental parameters such as larvae survival rate,pupation rate,eclosion rate and oviposition amount of treatment group was less than the control.5 The midgut and bady fat transcriptome of H.cunea was sequenced used high-throughput sequencing.We had 191,074,135 pieces clean reads and 93,571 Unigene,the average length of which is 839.83 bp.There are about 21,396 Unigene article can honology alignment to other species Nr databases.The midgut and bady fat transcriptome of H.cunea was analyzed.The 2,113 DEG was screened,of which 46 DEG was associated with five detoxification enzymes. |
