Philodendron 'con-go' is a kind of ornamental flower in Araceae.It is an excellent plant as afforestation in homes and public places.It is in great demand in the market.Tissue culture and rapid propagation is one of the main ways of seedling breeding of Philodendron 'con-go'.In this study,the effects of different sucrose,mannitol,growth inhibitors and culture medium components on the in vitro preservation of tissue cultured seedlings of Philodendron 'con-go ' were studied,the preserved tissue culture seedlings were restored to grow,and domestication and transplanting were carried out,a complete in vitro preservation system for tissue cultured seedlings of Philodendron 'con-go' was established.It provides technical support and theoretical basis for the large-scale production and popularization of tissue cultured seedlings of Philodendron 'con-go'.The main findings are as follows:1.Study on the slow growth and in vitro preservation of tissue cultured seedlings of Philodendron 'con-go'Add 0?5g/L sucrose,15?30g/L mannitol,the survival rate of Philodendron 'con-go' in vitro was high,which was 80%?90%and 70%?80.5%,respectively;Plant height changes little,SPAD values respectively were 33.32?43.86 and 37.47?20.77;the relative conductivity respectively were 33.19?35.40 and 33.21?34.16;the proliferation coefficients respectively were 3.67?3 and 8.67?12;the biomass of tissue culture seedlings increased under sucrose treatment and the biomass of tissue culture seedlings decreased under mannitol treatment.The microstructure of tissue culture seedlings is good.It is beneficial to the in vitro preservation of tissue culture seedlings of Philodendron 'con-go'.ABA treatment with 3?4mg/L showed good growth,the high survival rate respectively were 83.33%and 80%,the SPAD value were 57.34 and 58.88,the relative conductivity were 29.88 and 33.21,and the proliferation coefficient were 3.67 and 3.With 1.0?1.5mg/L PP333,the survival rate of tissue culture seedlings was 70%and 80.33%,growth potential is good,it was conducived to strong seedlings,the SPAD value were 58.08 and 60.90,the relative conductivity were 24.09 and 27.86,and the proliferation coefficient were 4 and 3.With the addition of 3?4mg/L B9,the survival rates of tissue culture seedlings respectively were 83.33%and 80%;growth potential was good,plant height changed little;the SPAD value were 57.34 and 58.88,the relative conductivity were 29.88 and 33.21,and the proliferation coefficient were 3.67 and 3.Under the treatment of growth inhibitor,the biomass of tissue cultured seedlings of Philodendron 'con-go' were all changed little,the microstructure of the leaves of the tissue culture seedlings were thickened gradually,which was beneficial to the preservation of tissue culture seedlings of Philodendron 'con-go' in vitro.Nutrient composition test of different medium,a large number of elements in MS medium were reduced,and the survival rate was lower,between 50%and 62.5%.The dead leaves of tissue culture seedlings were serious and the preservation effect was not good,which was not conducive to the in vitro preservation of tissue culture seedlings of Philodendron 'c on-go'.The correlation analysis of tissue culture seedlings of Philodendron 'con-go' showed that the plant height was negatively correlated with the proliferation coefficient(P<0.01),and the proliferation coefficient was negatively correlated with the SPAD value(P<0.01).It showed that the seedling height of tissue culture is inversely proportional to the proliferation coefficient.Through principal component analysis and membership function comprehensive evaluation,it was found that:in vitro preservation effect:B3>A4>P3>P2>B2>T0?T5>G15>CK>A3>G30.2.Study on the growth recovery of tissue cultured seedlings of Philodendron 'con-go' after in vitro preservation.In the tissue cultured seedlings of Philodendron'con-go' in vitro preservation restore growth after 60d,by compared the plant height,stem diameter,restore growth rate and the SPAD value and the tissue cultured seedlings aboveground biology characteristics,the rooting test of tissue culture seedlings was found:in vitro preservation of tissue cultured seedlings of Philodendron 'con-go' and contrast seedlings,there were no significant difference compared to restore growth rate of 100%;the POD and SOD activities of tissue cultured seedlings after in vitro presservation were gradually similar to those of the control seedlings,and there was no signifycant difference in biological characteristics between the recovered tissue cultured seedlings and the control seedlings.The genetic stability of the in vitro preserved tissue cultured seedlings is maintained,and the in vitro preservation method is feasible.3.Study on the domestication and transplanting of tissue cultured seedlings of Philodendron'con-go'after in vitro preservation.After in vitro preservation,the tissue culture seedlings of Philodendron 'con-go'were domesticated and transplanted,Biological characteristics of tissue culture seedlings preserved in vitro after transplan-ting,SPAD value and rooting ability were not significantly different from that of control tissue culture seedlings,the survival rate of transplanting is more than 90%,plant height,stem thick,leaf length,leaf width,leaf number and rooting were consistent with the control tissue culture seedlings.After in vitro preservation,the tissue culture seedlings of Philodendron 'con-go' can be restored to the normal growth state.
