Inner Mongolia cashmere goat is an excellent local breed in China,and the cashmere produced is a high-quality natural fiber textile raw material in the wool textile industry,which is known as "fiber gem","soft gold" and so on.The research group analyzed the production performance data of Inner Mongolia cashmere goat(Aerbas type).According to the coarse hair length,the Inner Mongolia cashmere goat was divided into three different hair types:long hair type(hair length>22cm),short hair type(hair length≤13 cm)and intermediate type(13cm<hair length≤22cm).It was found that coarse hair length has a certain reference value for the indirect selection of other important economic traits of cashmere,and the selection of long hair type can increase cashmere yield.Increase the length of cashmere and reduce the fineness of cashmere.In this study,based on the previous research of our group,72 skin tissue transcriptome sequencing data of long hair type and short hair type of Inner Mongolia cashmere goat in 12 months were analyzed by weighted gene co-expression network analysis of(WGCNA),to mine the co-expression modules related to different hair types,to mine candidate genes,and to detect the relative expression of key candidate genes and locate the protein expression.The main conclusions of the study are as follows:1.Based on the WGCNA analysis of 72 transcriptome data,the genes were divided into 19 co-expression modules,and it was found that there was a strong correlation between Magenta module and different hair types.There are 128 genes in this module,including 49 keratin related genes and several genes related to fat metabolism.GO functions are mainly concentrated in cellular components,including intermediate filaments,intermediate fiber cytoskeleton,cytoskeleton and keratin fibers.KEGG pathway is mainly enriched in MAPK signal pathway,PPAR signal pathway and arginine and proline metabolic pathway.Through screening,it was found that FGF21,FGF22,ASL,KRT25,KRT39,KRT74,KRTAP3-1,KRTAP11-1,TCHH,LOC102178266,LOC108634683 and LOC108637647 may be associated with different hair types of cashmere goats.2.The relative expressions of FGF21,ASL,KRT25,KRT39 and KRT74,were detected in the skin tissues of growing period(September),retrogression period(December)and resting period(March).The results showed that the expression levels of FGF21,ASL,KRT39 and KRT74 in long hair type were significantly higher than those in short hair type,and there was a very significant positive correlation between relative expression and hair length,and the correlation coefficient was between 0.75 and 0.83.The expression of KRT25 in the skin of long hair type was significantly higher than that of short hair type.It is speculated that these genes may be the key genes affecting different hair types of cashmere goats.3.FGF21 protein was localized in the skin of two different hair types of cashmere goats.The results showed that FGF21 was mainly expressed in primary hair follicle connective tissue sheath,primary hair follicle outer root sheath,secondary hair follicle outer root sheath,secondary hair follicle inner root sheath and sebaceous glands.The expression signal of FGF21 in primary hair follicles of long hair type is stronger than that of short hair type.It is speculated that FGF21 is related to the formation of hair length difference.
