The purpose of this study was to investigate the effects of roughage of different quality on rumen fermentation parameters,plasma biochemical parameters and rumen flora structure of dairy cows.8 Chinese Holstein dairy cows with similar weight,parity and milk productivity and in late lactation were assigned randomly to treatment and control groups.The roughage of control group were consisted by alfalfa silage,alfalfa and whole plant of corn silage,nameed CS group;the roughage of treatment groupwas consisted by straw,alfalfa silage,alfalfa and whole plant of corn silage,named RS group.The experiment lasted 21 days,including 14 days of pre-feeding and 7 days of formal sampling.The rumen fermentation parameters VFA,NH3-N,MCP,plasma biochemical indexes were measured and analyzed,rumen flora structure of dairy cows were sequenced and analyzed.The test results show that:Experiment 1:Effect of partial replacement of corn silage by rice straw on rumen fermentation of dairy cows:the main VFA concentrations of acetic acid,propionic acid and butyric acid in the rumen of the CS were significantly higher than RS(P<0.01);and NH3-N was higher than CS,but the difference was not significant(P>0.05).The MCP concentration.in the rumen of the CS was significantly lower than RS(P<0.05).Experiment 2:Effect of partial replacement of corn silage by rice straw on biochemical indexes of dairy cow plasma:the plasma TP,ALB,GLB,A/G of CS were not significantly different from RS(P>0.05),and BUN was significantly lower than RS(P<0.05);TC,TG,apo B-100,VLDL,GLU,LEP,and InS concentrations of the CS were also not significantly different from RS(P>0.05),but NEFA content of CS was significantly lower than RS(P<0.05);and the ALT,AST and CK activities of the CS were not significantly different from RS(P>0.05).Experiment 3:Effect of partial replacement of corn silage with rice straw on the structure of rumen microflora in dairy cows:at the phylum level,the relative abundance of Actinobacteria in CS was significantly lower than RS(P<0.01),and the relative abundance of Fibrobacteres and Firmicutes was significantly lower RS(P<0.05),Microsporidia,Blastocladiomycota,Cyanobacteria,Chytridiomycota,Cryptomycota,Ascomycota,Zoopagomycota,Mucoromycota,Spirochaetes and Basidiomycota were significantly higher than RS(P<0.05).At the genus level,the relative abundances of Butyrivibrio,Faecalibacterium,and Parabacteroides in the rumen of CS were significantly lower than RS(P<0.01),The relative abundances of Bacteroides,Alistipes,Oscillibacter,Fibrobacter were significantly lower than RS(P<0.05),Eubacterium,Neocallimastix,Spizellomyces,Piromyces,Rhizophagus,Anaeromyces,Gonapodya,Rhizoclosmatium and Sarcina were significantly higher than RS(P<0.05).In summary,the replacement of partial ratio of corn silage by rice straw has a significant effect on the rumen fermentation parameters of dairy cows and the structure of rumen bacterial bacterial.There were no significant differences in most of the plasma biochemical indicators,but the plasma NEFA concentration and BUN concentration increased significantly.
