Mapofloxacin(MBF)belongs to the fluoroquinolones.in order to clarify the safety of MBF in veterinary drug residue in broilers,HPLC method for detection of MBF in chicken plasma was established in this paper,and pharmacokinetic and residue tests of MBF raw drug and granular preparation(4.2%,denoted by MBF)were conducted in chickens.MBF HPLC test results showed that the standard curve had a good linearity in the range of 0.1 ?g /ml-10 ?g/ml,with an R2 value of 0.9998.The detection limit was 0.05 ?g/ml and the quantitative limit was 0.1 ?g/ml.Accuracy in and between batches was within ±15%.The results show that the HPLC method established in this experiment has high sensitivity,high accuracy and less interference.The stability under the conditions of 4-h room temperature,ultra-low temperature long-term preservation and repeated freezing-thawing meets the standard,which is suitable for the detection of MBF concentration in chicken plasma and tissues.Drug surrogate test of MBF in chickens: Broilers were given a dose of 2.5 mg/kg respectively in the form of bulk drug and granules(4.2% in form of MBF).After administered with MBF,blood was collected within 24 h and the blood concentration was determined by HPLC.The results showed that the pharmacokinetics of MBF in chickens was in line with the non-atrioventricular model,and its parameters were as follows: the peak time after gavage in the form of API was 1.92±0.66 h;The peak-reaching concentration was 0.82±0.2 ?g/ml.The area under the curve was 4.90±0.64 hr·?g/ml.The clearance half-life was 10.2±5.36 h.The average retention time was 4.62± 0.25 h.The clearance rate was 266±89 m L/hr/kg.The apparent volume of distribution was 3421±804 m L/kg.The peak time of granule(4.2%,in form of MBF)was 8.75±8.29 h after administration.The peak-reaching concentration was 3.31±0.53 ?g/ml.The area under the curve was 56.38±4.51 hr·?g/ml.The average retention time was 4.62± 0.25 h.The pharmacokinetics of MBF in broilers are characterized by large apparent volume of distribution,wide distribution in organs,long clearance half-life,and slow internal absorption.Based on the half-life results of 10.2h,it can be deduced that the gavage MBF of broilers is 102 h,about 4-5 days.The results of this experiment will provide a theoretical basis for the formulation of MBF during the broilers' drug rest period.MBF residue test: HPLC analysis was used to detect the residue of MBF in various tissues of broilers.The residue test results showed that MBF was mainly distributed in the heart,kidney and other tissues,and the concentration of MBF in these tissues was all lower than 30 g/kg,indicating that the residual amount of broilers 24 h after receiving MBF at 2.5 mg/kg dose met the MRL standard.However,after administered MBF Granule for 24 h,there were residues in the liver,heart,lung and kidney of broilers to varying degrees,among which the concentration of heart was higher than that of other tissues and the concentration of kidney was the lowest.Therefore,in the clinical application of MBF Granule,the dosage should be appropriately reduced to prevent residues and ensure the safety of animal-derived food.
