Vibrio anguillarum and Aeromonas salmonicida are two common pathogens in aquaculture,causing huge losses in aquaculture industry.Vaccination is one of the most effective measure to prevent various diseases.Taking into account of safety,efficacy and the cost,fish vaccines are still dominated by inactivated vaccines.However,adjuvants are often added into the inactivated vaccine owing to a low efficiency.Most commercial fish vaccines contains oil-adjuvant,but it causes serious side effects such as visceral adhesions,adjuvant residues and growth inhibition.Therefore,it's need to develop vaccines containing safe water-adjuvant urgently.Firstly,we constructed a pCpG adjuvant based on three CpG ODNs,using turbot as an experimental animal,and investigated the immunoprotection of the pCpG adjuvant combined with an inactivated V.anguillarum vaccine.The results showed that the pCpG adjuvant improved the immunoprotection of the inactivated vaccine,and the adjuvant dose was not positively related to the efficacy of the vaccine.At the same time,pCpG adjuvant effectively reduced the antigen dose of the inactivated vaccine,raised a high RPS of 84%and 43%increase compared with vaccine without pCpG.Immune response test suggested that pCpG increased the antibody level and activated innate and adaptive immune responses in turbot.This shows that pCpG adjuvant can be used as a candidate adjuvant for the development of fish vaccines.In addition,an inactivated vaccine against A.salmonicida was prepared based on an aluminum adjuvant and the immunoprotection of the vaccine was investigated.The results showed that the side effects of the aluminum adjuvant vaccine were significantly lower than the oil-adjuvaned vaccine,and stimulated the production of serum-specific antibodies in turbot.In summary,we discussed the application potential of two adjuvants with better safety than oil-adjuvant in fish vaccine development and provided a reference for screening safe and effective fish vaccine adjuvants.
